从8种树脂中筛选出能将γ-氨基丁酸与谷氨酸钠分离的最佳树脂,考察了色谱柱高径比、洗脱流速、柱温对分离度和回收率的影响,用γ-氨基丁酸发酵液上样,研究了发酵液的分离效果.结果表明,树脂QY-HG01能将γ-氨基丁酸与谷氨酸钠高效分离;在高径比25、流速2.5 m L/min、柱温50℃的条件下,γ-氨基丁酸与谷氨酸钠的分离效果最好;用预处理的γ-氨基丁酸发酵液上样,发酵液脱色率为84.9%,脱盐率为99.2%,γ-氨基丁酸的纯度由24.9%提高到84.0%,回收率为89.3%.
The optimum resin which could separate γ-aminobutyric acid and glutamic acid from eight kinds of resins was selected. The effects of column height-diameter ratio, elution flow rate and column temperature on the separation and recovery were investigated. - aminobutyric acid fermentation broth was used to study the separation efficiency of the fermentation broth.The results showed that the resin QY-HG01 could separate γ-aminobutyric acid and sodium glutamate efficiently.With the ratio of 25, the flow rate of 2.5 mL / min, the column temperature of 50 ℃ under the conditions, γ-aminobutyric acid and sodium glutamate separation of the best; pretreatment of γ-aminobutyric acid fermentation broth loading, the broth decolorization rate was 84.9%, the rate of desalination Is 99.2%, the purity of γ-aminobutyric acid is increased from 24.9% to 84.0%, and the recovery rate is 89.3%.
