目的:探讨酵母多糖对胰岛素抵抗(IR)小鼠糖脂代谢的影响。方法:8周龄C57BL/6J雄性小鼠根据体质量及基础血液生化指标随机分为4组,正常组给予标准饲料;模型组给予高脂纯化饲料喂养;阳性药小檗碱组及酵母多糖组给予高脂饮食喂养同时预防性给药,连续16周。检测血浆葡萄糖(GLU)、总胆固醇(TC)、甘油三酯(TG)、游离脂肪酸(FFA)、低密度脂蛋白胆固醇(LDL-C)等,进行口服糖耐量试验,ELISA法测血浆胰岛素并计算胰岛素抵抗指数HOMR-IR,解剖分离小鼠的肝脏和肌肉,测定组织中脂质含量,分离部分肝脏及附睾周围脂肪组织做HE染色进行形态学观察。结果:与模型组相比,小檗碱组和酵母多糖组的GLU、TC、TG、LDL-C含量均明显下降(P<0.01,P<0.05),血浆胰岛素水平极显著下降;酵母多糖组肝脏、肌肉中的TC、TG含量均显著下降(P<0.05)。结论:酵母多糖具有降低血糖、血脂,改善胰岛素抵抗的作用,是用于防治糖、脂代谢紊乱的有效物质,具有进一步研究的价值。
Objective: To investigate the effect of zymosan on glucose and lipid metabolism in insulin resistance (IR) mice. Methods: Eight-week-old C57BL / 6J male mice were randomly divided into 4 groups according to the body weight and basic blood biochemical parameters. The normal group was given standard diet. The model group was fed with high fat diet. The positive berberine group and zymosan group Give high-fat diet while feeding prophylactically for 16 weeks. Plasma glucose, total cholesterol, triglyceride, free fatty acid (FFA) and low density lipoprotein cholesterol (LDL-C) were measured for oral glucose tolerance test and plasma insulin The insulin resistance index HOMR-IR was calculated. The liver and muscle of mice were dissected. The lipids in the tissues were measured. The fatty tissues around the liver and epididymis were separated for HE staining and morphological observation. Results: Compared with model group, the content of GLU, TC, TG and LDL-C of berberine group and zymosan group decreased significantly (P <0.01, P <0.05), and the level of plasma insulin decreased significantly Liver and muscle TC, TG content were significantly decreased (P <0.05). Conclusion: Zymosan has the effects of lowering blood glucose, blood lipid and improving insulin resistance, and is an effective substance for preventing and treating the disorders of glucose and lipid metabolism. It has the value of further research.
