目的:探讨E-钙粘素(E-Cad)、基质金属蛋白酶-2(MMP-2)及其抑制剂(TIMP-2)表达与大肠癌浸润转移的关系。方法:采用S-P免疫组织化学染色技术,检测30例大肠腺癌,60例大肠癌组织E-Cad、MMP-2和TIMP-2的表达情况。结果:E-Cad的表达率在大肠腺瘤中为87.10%,显著高于大肠癌中的55.10%(P<0.05);其表达与大肠癌的大体类型有关,且随着大肠癌分化程度的降低而减少,与淋巴结转移呈负相关,E-Cad表达率越高患者的预后越好(P均<0.05)。MMP-2的表达率在大肠腺瘤中为26.67%,显著低于大肠癌中的86.67%(P<0.05);其表达与大肠癌的Dukes分期、分化程度、淋巴结转移和生存期均密切相关(P均<0.05)。TIMP-2的表达在大肠腺癌和大肠癌组织中没有显著性差异(P均>0.05),但其表达与大肠癌的Dukes分期、淋巴结转移、远隔脏器转移及生存期均有关(P均<0.05)。结论:E-Cad、MMP-2和TIMP-2的检测可以成为临床判断大肠癌的恶性程度、转移及预后的重要参考指标。
Objective: To investigate the relationship between E-cadherin, matrix metalloproteinase-2 (MMP-2) and its inhibitor (TIMP-2) and invasion and metastasis of colorectal cancer. Methods: The expressions of E-Cad, MMP-2 and TIMP-2 in 30 cases of colorectal adenocarcinoma and 60 cases of colorectal carcinoma were detected by S-P immunohistochemical staining. Results: The expression of E-Cad was 87.10% in colorectal adenoma, which was significantly higher than that in colorectal carcinoma (55.10%, P <0.05). The expression of E-cad was related to the general type of colorectal cancer. Decreased, and negatively correlated with lymph node metastasis. The higher the E-Cad expression rate, the better the prognosis of patients (all P <0.05). The expression of MMP-2 in colorectal adenoma was 26.67%, which was significantly lower than that in colorectal cancer (86.67%, P <0.05). The expression of MMP-2 was closely related to the Dukes stage, differentiation, lymph node metastasis and survival of colorectal cancer (P <0.05). TIMP-2 expression was not significantly different between colorectal adenocarcinoma and colorectal cancer (all P> 0.05), but the expression of TIMP-2 was related to Dukes stage, lymph node metastasis, distant organ metastasis and survival of colorectal cancer (P All <0.05). Conclusion: The detection of E-Cad, MMP-2 and TIMP-2 can be an important reference for clinical diagnosis of malignant degree, metastasis and prognosis of colorectal cancer.
