目的:探讨红花黄色素注射液治疗高原地区颅脑损伤大鼠的效果及对血浆降钙素基因相关肽(CGRP)及神经功能的影响。方法:选取120只SPF级成年雄性Wistar大鼠随机分为假手术组、模型组及红花注射液低、中、高剂量组,每组各24只,所有大鼠均于高原地区(4 300米海拔)饲养30 d,除假手术组外各组大鼠建立颅脑损伤模型,假手术组和模型组分别腹腔注射等体积生理盐水,红花黄色素注射液低、中、高剂量组分别腹腔注射红花黄色素注射液15、30、60 mg/kg,连续给药14 d后检测相关指标。结果:模型组在干预第3、7、14天血浆CGRP、SOD显著低于假手术组(P<0.05),红花黄色素注射液各剂量组血浆CGRP、SOD在干预第3、7、14天显著高于模型组(P<0.05);模型组在干预第3、7、14天血浆NSE、m NSS评分、神经细胞凋亡率及脑组织5-HT含量显著高于假手术组(P<0.05),红花黄色素注射液各剂量组血浆NSE、m NSS评分、神经细胞凋亡率及脑组织5-HT含量在干预第3、7、14天显著低于模型组(P<0.05)。其作用呈量效关系。结论:红花黄色素注射液对高原地区颅脑损伤大鼠具有显著的疗效,能显著增加血浆中CGRP含量,改善大鼠神经功能。
Objective: To investigate the effect of safflower yellow injection on traumatic brain injury in highland and its effect on plasma calcitonin gene related peptide (CGRP) and neurological function. Methods: A total of 120 SPF adult male Wistar rats were randomly divided into sham operation group, model group and Honghua injection low, medium and high dose groups, 24 rats in each group. All rats were in the plateau Rice elevation) for 30 days. Rats in each group except for the sham-operation group were given brain injury models. Results: The levels of plasma CGRP and SOD in model group were significantly lower than those in sham operation group on the 3rd, 7th and 14th day (P <0.05) (P <0.05). On the 3rd, 7th and 14th day after intervention, the plasma NSE and NSE scores, neuronal apoptosis rate and brain tissue 5-HT content in model group were significantly higher than those in sham operation group (P <0.05) (P <0.05). The plasma levels of NSE and NSE, the apoptotic rate of neurons and the content of 5-HT in brain tissue in each dose of safflower yellow injection group were significantly lower than those in model group on the 3rd, 7th and 14th days ). Its role was dose-effect relationship. Conclusion: Safflower yellow injection has a significant effect on rats with brain injury in plateau area, and can significantly increase the content of CGRP in plasma and improve the neurological function in rats.
