目的了解黑龙江省2014-2015年流感病毒优势株H3N2亚型HA基因变异特征。方法选取2014-2015年流感流行季节16株H3N2代表株进行HA基因扩增并测序,用DNAStar软件和MEGA 6软件分析序列并绘制基因进化树。结果 16株H3N2流感毒株经序列拼接后HA基因全长均为1701bp,编码566个氨基酸,同WHO推荐的北半球A/Switzerland/9715293/2013和A/Hong Kong/4801/2014 H3N2流感病毒疫苗株高度同源;黑龙江省从2015年11月开始H3N2亚型流感毒株逐渐由3C.3a向3C.2a支系转变,这与WHO推荐的2015-2016年流感病毒疫苗株A/Switzerland/9715293/2013(3C.3a)相比存在差别;16株H3N2代表株在抗原决定簇A区第142、第144位和B区第159、第160位上氨基酸发生改变。结论黑龙江省H3N2亚型流感病毒抗原性和HA1基因发生了一定的变异。
Objective To understand the variation of HA gene of influenza virus H3N2 subtype in Heilongjiang Province during 2014-2015. Methods 16 strains of H3N2 from influenza epidemic season in 2014-2015 were selected for HA gene amplification and sequencing. DNAStar software and MEGA 6 software were used to analyze the sequence and draw the phylogenetic tree. Results The total length of HA gene of 16 strains of H3N2 was 1701bp and encoded 566 amino acids. The results showed that the HA gene of 16 strains of H3N2 influenza virus was sequenced and stained with the same vaccine recommended by the WHO in the northern hemisphere A / Switzerland / 9715293/2013 and A / Hong Kong / 4801/2014 H3N2 influenza virus vaccine strain Highly homologous. From November 2015, the H3N2 subtype influenza strains gradually transformed from 3C.3a to 3C.2a in Heilongjiang Province. This is in line with the WHO recommended 2015-2016 influenza virus vaccine strain A / Switzerland / 9715293 / 2013 (3C.3a). The 16 strains of H3N2 changed their amino acids at positions 142 and 144 in region A and 159 and 160 in region B, respectively. Conclusion The antigenicity and HA1 gene of H3N2 influenza virus in Heilongjiang province have some variation.
