目的 :探讨慢性低O2 高CO2 脑神经细胞凋亡与MDA、NOS关系。方法 :将SD大鼠分为正常对照组(C组 )和低O2 高CO2 4周组 (E组 ) ,采用原位末端标记 (TUNEL)、流式细胞技术检测凋亡细胞 ,并测脑组织MDA、NOS水平。结果 :①E组脑MDA、NOS水平高于C组 (P <0 .0 1) ;②E组大脑皮质、海马、丘脑等部位见神经细胞凋亡 ;神经细胞凋亡百分率为 11.5 1% ,并与MDA和NOS水平呈正相关 (r =0 .6 5 2 ,P <0 .0 5和r=0 .716 ,P <0 .0 5 )。结论 :慢性低O2 高CO2 时氧自由基和一氧化氮生成增加是神经细胞凋亡形成的重要机制之一
Aims: To investigate the relationship between apoptosis and MDA and NOS in chronic hypoxic hypercapnic brain cells. Methods: SD rats were divided into normal control group (C group) and low O2 high CO2 4-week group (E group). TUNEL and flow cytometry were used to detect apoptotic cells and brain tissue MDA, NOS level. Results: ① The levels of MDA and NOS in E group were higher than those in C group (P <0.01). ② The apoptosis of neurons was found in cerebral cortex, hippocampus and thalamus in E group. The percentage of apoptosis was 11.5 1% MDA and NOS levels were positively correlated (r = 0.652, P <0.05 and r = 0.766, P <0.05). Conclusion: Increased production of oxygen free radicals and nitric oxide in chronic hypoxia and hypercapnia is one of the important mechanisms of neuronal apoptosis
