背景高血压及血压升高导致的血管损伤在人群中患病率很高,干细胞因子(SCF)及其受体在血管损伤修复方面有重要作用。SCF可分两型,可溶型SCF参与各种病理应激反应,膜结合型与慢性炎症时血管形成和纤维化有关。目的探讨外周血浆中干细胞因子(SCF)及其受体(c-kit)表达与血压水平之间的关系。方法采用横断面研究方法,随机入选156例作为研究对象,包括正常血压(n=36),高血压前期(n=45)和高血压病人(n=75)。测量腰围、血压、血脂、血糖等指标。用酶联免疫吸附法(ELISA)检测外周血浆中SCF及其受体蛋白的表达,放射免疫法检测内皮素1及肿瘤坏死因子α(TNF-α)水平。结果①SCF及c-kit水平随血压水平的升高而增加[SCF:正常血压(768.8±44.1)比高血压前期:(834.6±47.6)比高血压:(907.1±52.3)ng/L;c-kit:正常血压(9.6±1.5)比高血压前期(11.1±2.1)比高血压(13.2±1.6)mg/L,P<0.01];②SCF及c-kit表达水平与收缩压、舒张压、内皮素1和TNF-α水平呈正相关;③多元线性回归显示SCF、c-kit和内皮素1是收缩压的危险因素,SCF、c-kit、内皮素1和TNF-α是舒张压的危险因素。结论外周血中SCF及其受体c-kit表达水平随血压水平升高而有逐渐升高趋势,并与收缩压、舒张压、内皮素1和TNF-α水平呈正相关,提示高血压病人血管有炎症过程,内皮明显损伤。
BACKGROUND OF THE INVENTION Hypertension and high blood pressure lead to a high prevalence of vascular injury in humans. Stem cell factor (SCF) and its receptors play an important role in vascular injury repair. SCF can be divided into two types, soluble SCF involved in a variety of pathological stress response, membrane-bound and chronic inflammation of vascular formation and fibrosis related. Objective To investigate the relationship between the expression of stem cell factor (SCF) and its receptor (c-kit) and the blood pressure level in peripheral blood plasma. METHODS: A total of 156 patients were enrolled into the study as subjects, including normotensive (n = 36), prehypertensive (n = 45) and hypertensive patients (n = 75) using a cross-sectional study. Measuring waist circumference, blood pressure, blood lipids, blood glucose and other indicators. The expression of SCF and its receptor protein in peripheral blood was detected by enzyme-linked immunosorbent assay (ELISA). The levels of endothelin-1 and tumor necrosis factor-α (TNF-α) were detected by radioimmunoassay. Results ① The levels of SCF and c-kit were increased with the increase of blood pressure [SCF: normal blood pressure (768.8 ± 44.1) vs prehypertension: (834.6 ± 47.6), hypertension: (907.1 ± 52.3) ng / L; kit: The normal blood pressure (9.6 ± 1.5) was higher than that of prehypertension (11.1 ± 2.1) and hypertension (13.2 ± 1.6) mg / L, P <0.01]; ②The expression of SCF and c-kit was correlated with systolic blood pressure, ③The multivariate linear regression showed that SCF, c-kit and endothelin-1 were the risk factors for systolic blood pressure. SCF, c-kit, endothelin-1 and TNF-α were the risk factors of diastolic blood pressure . Conclusions The expression of SCF and its receptor c-kit in peripheral blood increases gradually with the increase of blood pressure, and is positively correlated with the systolic pressure, diastolic pressure, the level of endothelin-1 and TNF- Inflammation process, endothelial damage.
