目的探讨白细胞介素5(interleukin-5,IL-5)和核因子κB(nuclear factor kappaB,NF-κB)在变应性鼻炎动物模型骨髓中表达的意义。方法采用6-8周雄性SD大鼠,随机分成实验组10只和对照组10只,以卵清蛋白致敏激发制成变应性鼻炎模型,免疫组化染色检测骨髓中IL-5和NF-κB的表达。结果大鼠变应性鼻炎模型骨髓涂片中IL-5阳性细胞和NF-κB比例都显著高于对照组(P<0.05),从形态上判断IL-5阳性细胞和NF-κB阳性细胞是嗜酸性粒细胞,IL-5阳性细胞比例和NF-κB阳性细胞比例呈显著正相关(r=0.79,P<0.01)。结论大鼠变应性鼻炎动物模型骨髓原位嗜酸性粒细胞能产生IL-5和NF-κB,可能调节骨髓中嗜酸性粒细胞的增殖和分化。
Objective To investigate the expression of interleukin-5 (IL-5) and nuclear factor kappaB (NF-κB) in the bone marrow of an allergic rhinitis animal model. Methods Male Sprague-Dawley rats aged 6-8 weeks were randomly divided into experimental group (n = 10) and control group (n = 10). Allergic ovalbumin-induced allergic rhinitis model was made. Immunohistochemical staining was used to detect the expression of IL-5 and NF -κB expression. Results The proportion of IL-5 positive cells and NF-κB in the bone marrow smear of allergic rhinitis model rats were significantly higher than those in the control group (P <0.05). The morphology of IL-5 positive cells and NF-κB positive cells were There was a significant positive correlation between the proportion of eosinophils and IL-5 positive cells and the percentage of NF-κB positive cells (r = 0.79, P <0.01). CONCLUSION: In situ eosinophils can produce IL-5 and NF-κB in the rat model of allergic rhinitis, which may regulate the proliferation and differentiation of eosinophils in bone marrow.
