目的研究CD44st和基质金属蛋白酶2(MMP-2)在非小细胞肺癌(NSCLC)组织中的表达及临床意义。方法采用荧光定量PCR和免疫组化法分别检测106例NSCLC组织及其30例对应的癌旁组织中CD44st和MMP-2mRNA和蛋白表达,分析其与临床病理学特征的关系。结果 NSCLC组织中CD44st和MMP-2mRNA和蛋白表达均高于癌旁组织(P<0.01)。CD44st与MMP-2mRNA表达呈线性相关(r=0.966,P<0.01)。CD44st和MMP-2 mRNA表达与患者组织病理类型、淋巴结转移及TNM分期密切相关(P<0.05),而与性别、年龄、分化程度及肿瘤大小无关(P>0.05)。CD44st和MMP-2高表达患者术后2年无疾病进展生存率均低于低表达患者(P<0.05)。结论 CD44st和MMP-2在NSCLC组织中高表达,在NSCLC进展中发挥重要作用,可能成为NSCLC预后判断和治疗的新靶点。
Objective To study the expression and clinical significance of CD44st and MMP-2 in non-small cell lung cancer (NSCLC). Methods The mRNA and protein expressions of CD44st and MMP-2 in 106 NSCLC tissues and 30 corresponding paracancerous tissues were detected by real-time fluorescence quantitative PCR and immunohistochemistry. The relationship between them was analyzed with clinicopathological features. Results The expressions of CD44st and MMP-2 mRNA and protein in NSCLC tissues were significantly higher than those in adjacent non-cancerous tissues (P <0.01). The expression of CD44st and MMP-2 mRNA showed a linear correlation (r = 0.966, P <0.01). The expressions of CD44st and MMP-2 mRNA were closely related to histopathological types, lymph node metastasis and TNM stage (P <0.05), but not to gender, age, differentiation and tumor size (P> 0.05). Patients with high expression of CD44st and MMP-2 had no disease progression at 2 years after operation compared with patients with low expression (P <0.05). Conclusion The high expression of CD44st and MMP-2 in NSCLC tissues plays an important role in the progression of NSCLC and may be a new target for the prognosis and treatment of NSCLC.
