目的:分析银线莲水提物中的胞苷、尿苷、腺嘌呤、肌苷、鸟苷、腺苷6种核苷类物质。方法:高效液相色谱法-电喷雾电离/离子阱质谱法(HPLC-ESI/MS)对银线莲中超声波辅助萃取法水提物中6种核苷类物质进行测定和鉴别。结果:银线莲药材中检测到胞苷、尿苷、腺嘌呤、肌苷、鸟苷和腺苷6种核苷成分。6种核苷类物质质量浓度在1.25~80.0μg·ml-1范围内呈良好线性关系(r≥0.999 4),回收率在92.7%~96.8%之间,RSD≤1.6%。测得银线莲药材提取物中胞苷、尿苷、腺嘌呤、肌苷、鸟苷和腺苷含量分别为0.106 2,0.339 1,0.069 9,0.082 0,0.030 8,0.107 4 mg·g-1。结论:此法简便、灵敏快速,首次报道银线莲中含有重要药理活性的6种核苷类物质,为银线莲的药理和临床试验研究提供理论基础。
OBJECTIVE: To analyze six nucleosides, cytidine, uridine, adenine, inosine, guanosine and adenosine in the aqueous extract of Anemone. Methods: Six kinds of nucleosides in aqueous extract by ultrasonic-assisted extraction were determined and identified by HPLC-ESI / MS. Results: Cytosine, uridine, adenine, inosine, guanosine and adenosine were detected in Anemone. The linear range of the six nucleosides was 1.25 ~ 80.0μg · ml-1 (r≥0.9994). The recoveries ranged from 92.7% to 96.8% with a RSD ≤1.6%. The contents of cytidine, uridine, adenine, inosine, guanosine and adenosine in the extracts of A. purpurea were 0.106, 2.0339, 1.069 9, 0.082 0, 0.030 8, 0.107 4 mg · g- 1. CONCLUSION: This method is simple, sensitive and rapid. It reports for the first time the six nucleosides containing important pharmacological activities in Anemarrhena asphodeloides, providing a theoretical basis for the pharmacological and clinical trials of Anemones.
