目的探讨美洲大蠊提取物对人肺癌细胞H125影响及其作用机制。方法将H125细胞分为阴性对照组、阳性对照组(顺铂10μg/mL)、美洲大蠊提取物(10、20、40μg/mL)组,四甲基偶氮噻唑蓝比色法观察H125细胞生长状况,流式细胞仪分析细胞周期、细胞凋亡及线粒体膜电位,蛋白免疫印迹法检测细胞中Bax、Bcl-2蛋白的表达。结果美洲大蠊提取物可以抑制H125细胞的生长,IC50为24.8μg/mL;美洲大蠊提取物可明显降低H125细胞的线粒体膜电位,与对照组比较,40μg/mL美洲大蠊组H125细胞绿色荧光细胞的百分率[(34.12±0.21)%]及凋亡率[(24.76±0.168)%]升高,同时G2/M期细胞比例[(23.12±0.159)%]减少,S期细胞比例[(20.92±0.096)%]增加,Bax蛋白表达增强,Bcl-2蛋白表达受到明显抑制。结论美洲大蠊提取物可以抑制人肺癌细胞H125的生长,阻滞细胞周期于S期,并通过线粒体途径诱导细胞凋亡。
Objective To investigate the effect of Periplaneta americana extract on human lung cancer cell line H125 and its mechanism. Methods H125 cells were divided into negative control group, positive control group (cisplatin 10μg / mL), Periplaneta americana extract (10,20,40μg / mL) group, tetramethyl azothiazole blue colorimetric assay H125 cells Growth status, cell cycle, apoptosis and mitochondrial membrane potential were analyzed by flow cytometry. The protein expression of Bax and Bcl-2 were detected by Western blot. Results Periplaneta americana extract could inhibit the growth of H125 cells with an IC50 of 24.8 μg / mL. Periplaneta americana extract could significantly reduce the mitochondrial membrane potential of H125 cells. Compared with the control group, the H125 cells in 40 μg / mL American cockroach group had a green (34.12 ± 0.21)%] and apoptotic rate [(24.76 ± 0.168)%], while the proportion of cells in G2 / M phase [(23.12 ± 0.159)%] decreased while the percentage of cells in S phase [( 20.92 ± 0.096)%] increased, the expression of Bax protein increased, the expression of Bcl-2 protein was significantly inhibited. Conclusion Periplaneta americana extract can inhibit the growth of human lung cancer cell line H125, arrest the cell cycle in S phase and induce apoptosis through mitochondrial pathway.
