半薄切片(Semi thin section)介于石腊切片和超薄切片之间,是连接细胞微观与超微观结构的重要技术环节。这种切片厚度在0.5—1.0微米之间。由于细胞成份保存良好, 受色反应优异而具有光学高分辨性,在油镜下呈现类似低倍电镜的图像。因此,半薄切片已广泛用于细胞学和病理学的研究,制备精确的光镜照片和超微结构的定位观察。半薄切片一般是用超薄片机制备。但是,这种仪器价格昂贵,不能普及,限制了半薄切片的广泛开展。1964年,徐国明
Semi-thin section (Semi thin section) Between the paraffin section and the ultra-thin sections, is an important technology link cell microstructure and ultrastructure. The slice thickness is between 0.5-1.0 microns. Due to the well-preserved cell components, excellent color reaction and optical high resolution, the oil microscopic images appear similar to low power electron microscopy. Therefore, the semi-thin section has been widely used in cytology and pathology research, preparation of accurate light microscopy and ultrastructure of the positioning observation.
