目的 研究乳腺非典型增生及癌变过程中PCNA及癌基因蛋白p53、c-erbB-2的变化规律,并探讨在此过程中细胞增殖活性与p53、c-erbB-2癌基因蛋白表达的关系。方法 应用免疫组化法分析了86例良恶性乳腺组织中的PCNA,癌基因蛋白p53、c-erbB-2表达及其相互关系。结果PCNA、p53、c-erbB-2的表达随非典型增生程度的增加而增多。p53、c-erbB-2阳性表达病例PCNCN-LI明显高子阴性病例(t=2.9040,P<0.01;t=3.2333,P<0.01)。结论 PCNA、p53、c-erbB-2可反映乳腺癌前病变的恶变倾向,并对癌前病变的临床处理和随访有指导作用;细胞增殖活性与r53、c-erbB-2基因激活,蛋白产物聚积有密切联系。
Objective To study the changes of PCNA and oncogene proteins p53 and c-erbB-2 in the process of atypical hyperplasia and carcinogenesis of breast, and to explore the relationship between the cell proliferation activity and the expression of p53 and c-erbB-2 oncogene proteins in this process. Methods The expression of PCNA, oncogene proteins p53 and c-erbB-2 in 86 benign and malignant breast tissues were analyzed by immunohistochemistry. Results The expression of PCNA, p53 and c-erbB-2 increased with the increase of atypical hyperplasia. PCNCN-LI was significantly higher in patients with positive p53 and c-erbB-2 expressions (t=2.9040, P<0.01; t=3.2333, P<0.01). Conclusion PCNA, p53 and c-erbB-2 can reflect the malignant tendency of precancerous lesions of breast cancer, and guide the clinical treatment and follow-up of precancerous lesions; cell proliferation activity and r53, c-erbB-2 gene activation, protein production Accumulation is closely linked.
