目的:探讨缺氧与梭曼中毒单一或复合作用对培养大鼠心肌细胞的损伤特点和可能机制。方法:培养的大鼠心肌细胞在缺氧条件下暴露于含不同浓度梭曼的培养液中,观察细胞存活率和培养液肌酸磷酸激酶(CPK)、乳酸脱氢酶(LDH)及谷草转氨酶(GOT)等心肌酶含量变化,同时用Fura-2/AM荧光负载测定心肌细胞内游离钙浓度([Ca2+]i)变化。结果:缺氧与梭曼中毒均可导致培养的大鼠心肌细胞存活率下降,培养液中心肌酶含量升高,同时心肌细胞内游离钙浓度也明显增加;缺氧和梭曼中毒两种致伤因素同时作用对培养的大鼠心肌细胞所产生的复合效应显著大于单一因素的致伤结果。结论:缺氧可以加重梭曼对心肌细胞的毒性;缺氧或梭曼中毒所致心肌细胞的损伤可能与心肌细胞内钙超载有关
Objective: To investigate the characteristics and possible mechanisms of single or combined effects of hypoxia and soman intoxication on cultured rat cardiomyocytes. Methods: Cultured rat cardiomyocytes were exposed to soman medium containing different concentrations of hypoxia. The survival rate of cells and the activity of creatine phosphokinase (CPK), lactate dehydrogenase (LDH) and aspartate aminotransferase (GOT), and the changes of intracellular free calcium concentration ([Ca2 +] i) were measured by Fura-2 / AM fluorescence load. RESULTS: Both hypoxia and soman intoxication could lead to the decrease of the survival rate of cardiomyocytes, the increase of myocardial enzyme concentration in the culture medium and the increase of the intracellular free calcium concentration in the culture medium. Conclusion: Hypoxia may aggravate the cytotoxicity of soman to cardiomyocytes. The damage of cardiomyocytes induced by hypoxia or soman may be related to the overload of intracellular calcium
