目的:研究二氢槲皮素对异丙肾上腺素致大鼠心肌缺血的保护作用,探讨其对冠心病的防治作用及机制。方法:将SD大鼠随机分为空白组、模型对照组、阳性对照组(地奥心血康42 mg.kg-1)、二氢槲皮素高、中、低剂量组(6.4,3.2,1.6mg.kg-1)。皮下注射5 mg.kg-1异丙肾上腺素(ISO)造成心肌缺血性损伤模型后观察心电图(ECG)的变化,检测血清和心肌组织中超氧化物歧化酶(SOD)、丙二醛(MDA)和血清中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)和乳酸脱氢酶(LDH)含量变化,并对大鼠心肌组织进行了病理切片检查。结果:二氢槲皮素能改善大鼠ECG的J点位移,显著降低心肌缺血大鼠血清中CK,CK-MB,LDH的含量,升高心肌组织匀浆中SOD活力,降低心肌组织匀浆中脂质过氧化终产物MDA的含量。结论:二氢槲皮素能有效抑制ISO所致心肌组织缺血,保护心肌细胞免受损伤,从而起到治疗冠心病的作用。
Objective: To study the protective effect of dihydroquercetin on isoproterenol-induced myocardial ischemia in rats and its preventive and therapeutic effects on coronary heart disease. Methods: SD rats were randomly divided into blank group, model control group, positive control group (Dioxintangxunkan 42 mg.kg-1), dihydroquercetin high, medium and low dose groups (6.4,3.2,1.6mg.kg -1). The changes of electrocardiogram (ECG) were observed after subcutaneous injection of 5 mg.kg-1 isoproterenol (ISO) into the myocardial ischemic injury model. The activities of superoxide dismutase (SOD), malondialdehyde (MDA) ), Serum creatine kinase (CK), creatine kinase (CK-MB) and lactate dehydrogenase (LDH) were detected by enzyme-linked immunosorbent assay (ELISA). Results: Dihydroquercetin could improve the J-point displacement of ECG in rats and significantly reduce the contents of CK, CK-MB and LDH in the serum of myocardial ischemia rats, increase the activity of SOD in myocardial homogenate and decrease the myocardial homogenate Pulp content of lipid peroxidation final product MDA. Conclusion: Dihydroquercetin can effectively inhibit myocardial ischemia induced by ISO and protect cardiomyocytes against injury, thus playing a role in the treatment of coronary heart disease.
