目的 探讨原发性小肝癌 (SHCC)切除术后血清肝细胞生长因子 (HGF)水平和癌组织中c Met受体表达与术后复发的关系。方法 采用ELISA对手术前后血清HGF浓度变化进行检测 ,并利用Western、Northern印迹杂交方法 ,测定肿瘤与非肿瘤组织内c Met蛋白表达量。结果 术前血清HGF浓度为 0 64± 0 15ng/ml,术后 3天为 1 41±0 2 8ng/ml,二者差异有显著性意义 (P <0 0 0 1)。术后 3天血清HGF浓度是术前的 2 3 2± 0 5 5倍。术后HGF的增长倍数与肿瘤组织内c Met蛋白的过度表达在术后复发与未复发者中差别有显著性意义(P <0 0 5 ) ,两年内复发者明显高于两年后复发及未复发者。结论 SHCC切除术后血清HGF水平明显增高 ,肿瘤组织本身c Met蛋白过度表达 ,可能与SHCC术后早期复发有关
Objective To investigate the relationship between the level of serum hepatocyte growth factor (HGF) and the expression of c-Met receptor in post-operative recurrence of primary small hepatocellular carcinoma (SHCC). Methods The changes of serum HGF concentration before and after operation were detected by ELISA. The expression of c-Met protein in tumor and non-tumor tissues was detected by Western blot and Northern blotting. Results The preoperative serum HGF concentration was 0 64 ± 0 15 ng / ml, and the postoperative 3 days was 1 41 ± 0 2 8 ng / ml. There was a significant difference between the two groups (P <0.01). The serum HGF concentration 3 days after operation was 23 2 ± 0 5 5 times before surgery. There was a significant difference between postoperative HGF growth rate and c Met protein overexpression in recurrence and non-recurrence (P <0.05), and recurrence in two years was significantly higher than that in two years No recurrence. Conclusions After HCC, the level of serum HGF is obviously increased, the c Met protein overexpression in the tumor tissue may be related to the early postoperative recurrence of SHCC
