目的:比较马钱子碱复合磷脂脂质体[由大豆磷脂(SPC)、氢化大豆磷脂(HSPC)2种磷脂组成]和相应的2种单一磷脂(SPC、HSPC)脂质体的体外稳定性。方法:采用硫酸铵梯度法制备了马钱子碱脂质体,比较了不同磷脂组成马钱子碱脂质体的包封率、粒径、泄漏率、在血清中的稳定性和放样稳定性等性质。结果:在药脂摩尔比为0.14时,磷脂组成对马钱子碱脂质体的包封率基本没有影响。马钱子碱复合磷脂脂质体的平均粒径为159nm,明显高于相应的单一磷脂脂质体(108~113nm)。马钱子碱复合磷脂脂质体的稳定性介于HSPC脂质体和SPC脂质体之间。马钱子碱HSPC脂质体存在一个明显的突释过程。结论:体外稳定性实验结果表明,马钱子碱复合磷脂脂质体与相应的单一磷脂脂质体相比更具有研究开发的前景。
OBJECTIVE: To compare the in vitro stability of strychnine complex phospholipid liposomes (composed of two kinds of phospholipids of soybean phospholipid (SPC) and hydrogenated soybean phospholipid (HSPC)] and the corresponding two kinds of single phospholipids (SPC, HSPC) liposomes . Methods: The strychnine liposomes were prepared by ammonium sulfate gradient method. The encapsulation efficiency, particle size, leakage rate, serum stability and loft stability of strychnine liposomes with different phospholipid compositions were compared Other properties. Results: When the molar ratio of lipid to lipid was 0.14, the encapsulation efficiency of strychnine liposomes had no effect on the composition of phospholipids. The average size of the strychnine complex phospholipid liposomes was 159 nm, significantly higher than the corresponding single phospholipid liposomes (108 ~ 113 nm). The stability of strychnine complex phospholipid liposomes is between that of HSPC liposomes and SPC liposomes. Brucine HSPC liposomes have a significant burst process. Conclusion: The in vitro stability of the experimental results show that the strychnine phospholipid liposomes with the corresponding single phospholipid liposome more than the research and development prospects.
