目的:研究安胃丸对慢性胃炎大鼠血清炎性细胞因子及胃黏膜核因子-κB p65 mRNA(NF-κB p65 mRNA)表达的影响。方法:采用ig60%乙醇,20 mmol.L-1去氧胆酸钠ig,0.05%氨水自由饮用,12周复制大鼠慢性胃炎模型,将造模成功大鼠分为模型组、安胃丸低、中、高组。安胃丸低、中、高组分别以3.06,6.12,12.24 g.kg-1.d-1 ig给药,连续给药30 d,末次给药后禁食16 h取血,运用ELISA法检测给药后大鼠血清白介素-2(IL-2)、白介素-4(IL-4)、肿瘤坏死因子-α(TNF-α)含量变化,用原位杂交法检测大鼠胃黏膜核因子-κB p65 mRNA的表达情况。结果:安胃丸组胃黏膜上皮细胞排列整齐,腺体无萎缩现象;安胃丸组大鼠血清IL-4和IL-2水平分别高于、低于模型组,TNF-α水平较模型组显著降低,高、中剂量组大鼠胃窦黏膜NF-κB p65 mRNA的表达低于模型组,接近正常对照组。结论:安胃丸具有保护胃黏膜、减轻黏膜炎症反应的作用,其作用可能与抑制NF-κB p65 mRNA表达,减少炎性因子产生及调节机体免疫功能,维持体液免疫和细胞免疫平衡状态有关。
Objective: To study the effect of Angu Pills on serum inflammatory cytokines and the expression of NF-κB p65 mRNA in chronic gastritis rats. Methods: Chronic gastritis model was established by ig 60% ethanol, 20 mmol.L-1 deoxycholate sodium ig and 0.05% ammonia water for 12 weeks. The successful model rats were divided into model group, Anshu pill , Middle and high group. Anwei pills low, medium and high groups were given 3.06,6.12,12.24 g.kg-1.d-1 ig administration, continuous administration of 30 d, after the last administration fasting 16 h blood, the use of ELISA assay After administration, the contents of IL-2, IL-4 and TNF-α in serum of rats were detected by in situ hybridization. The levels of serum interleukin- κB p65 mRNA expression. Results: The gastric mucosal epithelial cells in Anweiwan group were arranged neatly with no shrinkage of gland. The serum levels of IL-4 and IL-2 in Anguis Pill group were higher than those in model group and the levels of TNF-α in model group The expression of NF-κB p65 mRNA in antral mucosa of rats in high and middle dose groups was significantly lower than that in model group and close to normal control group. Conclusion: Anguisin can protect the gastric mucosa and reduce the inflammatory reaction of mucosa, which may be related to inhibiting the expression of NF-κB p65 mRNA, decreasing the production of inflammatory cytokines, regulating the immune function and maintaining humoral and cellular immune balance.
