目的探讨人宫颈癌组织中胰岛素样生长因子-Ⅱ(IGF-Ⅱ)和H19印记缺失(LOI)与人乳头瘤病毒(HPV)感染的关系。方法2004-01-2004-12西安交通大学医学院第一附属医院采用逆转录-聚合酶链反应(RT-PCR)检测40例宫颈癌组织及20例正常宫颈组织中IGF-Ⅱ及H19的印记状态;聚合酶链反应(PCR)检查宫颈癌和对照组HPV16,18感染情况。结果宫颈癌组织中IGF-Ⅱ和H19的LOI(47.6%,26.3%)高于正常宫颈组织LOI(7.7%,0)(P<0.05)。在IGF-Ⅱ和H19的LOI肿瘤中,HPV感染率分别为80.0%和100.0%,高于宫颈癌的总体感染率和对照组的感染率(分别为65.0%和5.0%)(P<0.05)。结论IGF-Ⅱ和H19的LOI与宫颈癌发生有关。IGF-Ⅱ和H19的LOI可能与HPV感染有关。
Objective To investigate the relationship between insulin-like growth factor-Ⅱ (IGF-Ⅱ) and H19 imprinting deletion (LOI) and human papillomavirus (HPV) infection in human cervical cancer. Methods The first Affiliated Hospital of Xi’an Jiaotong University School of Medicine from January 2004 to December 2004 detected the expression of IGF-Ⅱ and H19 in 40 cases of cervical cancer and 20 cases of normal cervical tissue by reverse transcription-polymerase chain reaction (RT-PCR) Status; HPV16,18 infection in cervical cancer and control group by polymerase chain reaction (PCR). Results The LOI (47.6%, 26.3%) of IGF-Ⅱ and H19 in cervical cancer was significantly higher than that of normal cervical tissue (7.7%, 0) (P <0.05). In the LOI tumors of IGF-II and H19, HPV infection rates were 80.0% and 100.0%, respectively, higher than the overall infection rate of cervical cancer and the control group (65.0% and 5.0% respectively) (P <0.05) . Conclusion The LOI of IGF-Ⅱ and H19 is related to the occurrence of cervical cancer. The LOI of IGF-II and H19 may be related to HPV infection.
