目的观察苓桂术甘汤(Linggui Zhugan decoction,LGZGD)含药血清对转化生长因子-β_1(Transforming growth factor,TGF-β_1)诱导的大鼠心肌细胞H9c2损伤的保护作用。方法分别使用10%的空白大鼠血清、LGZGD小、中、大剂量组含药血清和大鼠心肌细胞H9c2共培养,12 h后加入20 ng·m L~(-1)的TGF-β_1建立大鼠心肌细胞损伤模型,继续培养12 h后采用ELISA法检测细胞中TNF-α、IL-6和IL~(-1)β的含量。结果与正常组比,TGF-β_1组TNF-α、IL-6和IL~(-1)β的含量明显升高(P<0.01),与TGF-β_1组比,LGZGD 4.2,8.4 g·kg~(-1)组的TNF-α的含量显著下降,分别由(323.839±35.614)下降至(191.399±43.203)和(175.030±19.589)ng·L~(-1)(P<0.05、P<0.01);LGZGD 4.2,8.4g·kg~(-1)组的IL-6的含量由(67.171±13.506)下降至(40.490±7.899)(30.548±2.226)ng·L~(-1)(P<0.05、P<0.01),LGZGD2.1,4.2,8.4 g·kg~(-1)组的IL~(-1)β含量由(23.463±3.053)下降至(14.793±4.244)、(14.600±3.517)和(9.253±2.720)ng·L~(-1)(P<0.05、P<0.01)。结论 LGZGD含药血清对TGF-β_1诱导的大鼠心肌细胞H9c2的损伤有显著的保护作用。
Objective To observe the protective effects of Linggui Zhugan Decoction (LGZGD) -containing serum on H9c2 injury induced by transforming growth factor-β1 (TGF-β1) in rat cardiomyocytes. Methods 10% blank rat serum, LGZGD small, medium and large doses of drug-containing serum and rat cardiac myocytes H9c2 co-culture, after 12 h by adding 20 ng · m L -1 of TGF-β 1 established After the cells were cultured for 12 h, the levels of TNF-α, IL-6 and IL-1β in the rat myocardial cells were detected by ELISA. Results Compared with normal group, the levels of TNF-α, IL-6 and IL-1β in TGF-β1 group were significantly increased (P <0.01) (P <0.05), and the levels of TNF-αin the group were significantly decreased from (323.839 ± 35.614) to (191.399 ± 43.203) and (175.030 ± 19.589) ng · L -1, respectively 0.01). The levels of IL-6 in LGZGD 4.2, 8.4 g · kg -1 group decreased from (67.171 ± 13.506) to (40.490 ± 7.899) (30.548 ± 2.226) ng · L -1 (P <0.01, P <0.01). Conclusion The serum containing LGZGD can significantly protect H9c2 induced by TGF-β1 in rat cardiomyocytes.
