目的探讨中国东南沿海地区人群apelin及其受体APJ基因的单核苷酸多态性(SNP)与高血压的相关性。方法来自横断面调查研究对象共1031人,其中高血压组556例,对照组475人。Taqman@MGB探针法获得apelin基因rs3115757、rs56204867、rs3761581和APJ基因rs7119375、rs9943582共5个位点的基因分型。比较高血压组和对照组各个基因型分布差异,应用Logistic回归分析apelin和APJ基因5个SNP与高血压的关系。结果男性apelin基因3个位点突变等位基因频率在高血压组和对照组间分布差异有统计学意义(均P<0.05);APJ基因2个位点突变等位基因频率在高血压组和对照组间分布差异无统计学意义(均P>0.05)。女性的5个位点突变等位基因频率在高血压组和对照组间分布差异无统计学意义(均P>0.05)。调整年龄、体质量指数、空腹血糖后,Logistic回归分析显示,无论男性还是女性,携带rs3115757-C、rs56204867-C、rs3761581-A突变等位基因者分别较携带rs3115757-G、rs56204867-G、rs3761581-T者患高血压的风险增加(男性OR=6.34、2.41、1.95,女性OR=2.07、2.05、2.00,均P<0.05)。高血压最佳基因-基因作用模型男、女均为apelin rs3761581-apelin rs3115757-APJ rs7119375模式。结论本研究人群apelin基因的3个SNP rs3115757、rs56204867、rs3761581及APJ基因rs7119375与高血压有关。
Objective To investigate the association between apelin and its receptor APJ gene SNP and hypertension in the southeastern coastal areas in China. Methods From the cross-sectional survey of a total of 1031 people, including hypertensive group 556 cases, 475 control group. Taqman @ MGB probe method to obtain apelin gene rs3115757, rs56204867, rs3761581 and APJ gene rs7119375, rs9943582 a total of five loci genotyping. Logistic regression analysis was used to analyze the relationship between 5 SNPs of apelin and APJ gene and hypertension in hypertensive group and control group. Results There was significant difference in allele frequencies of three apelin gene mutations between hypertensive group and control group (all P <0.05). The frequencies of two alleles in APJ gene were significantly higher in hypertension group and There was no significant difference in the distribution among the control groups (all P> 0.05). There were no significant differences in allele frequencies among the five female sites in hypertension group and control group (all P> 0.05). After adjustment for age, body mass index and fasting blood glucose, logistic regression analysis showed that rs3115757-C, rs56204867-C and rs3761581-A alleles were significantly higher than those carrying rs3115757-G, rs56204867-G and rs3761581 -T patients with increased risk of hypertension (male OR = 6.34,2.41,1.95, female OR = 2.07,2.05,2.00, all P <0.05). The best model of hypertension gene-gene model male and female apelin rs3761581-apelin rs3115757-APJ rs7119375 model. Conclusion The three SNP rs3115757, rs56204867, rs3761581 and APJ rs7119375 of apelin gene in this study population are associated with hypertension.
