为评估hApoCⅢ转基因小型猪的环境生物安全性,采用PCR技术,通过检测其粪便、接触的土壤、肠道菌、代孕母猪的血液DNA以及与之接触的普通猪DNA是否发生了水平基因转移。同时通过大鼠90d经口毒性试验,检测大鼠的血液、生理生化指标和脏器系数是否发生了显著性变化。结果显示:与对照组相比,hApoCⅢ转基因小型猪的肠道内微生物菌群、普通猪的血液和代孕母猪的血液、周围环境土壤都没有发生基因水平转移现象。与对照组相比,试验组大鼠各项观察指标及病理检测均正常,血液、生化指标均在正常范围;初步显示hApoCⅢ转基因小型猪对生物环境是安全的,没有对环境造成不利影响。
To assess the environmental safety of hApoCⅢ transgenic miniature pigs, PCR techniques were used to detect the level of gene transfer by examining their faeces, soil contacted, gut bacteria, and sows’ blood DNA and the normal porcine DNA in contact therewith. At the same time, rats were subjected to oral toxicity test for 90 days to detect whether there was a significant change in blood, physiological and biochemical indexes and organ coefficient in rats. The results showed that compared with the control group, the intestinal microbiota of hApoC Ⅲ transgenic piglets, the blood of normal pigs and the blood of the surrogate sows and the surrounding soil did not undergo gene level transfer. Compared with the control group, all the observation and pathological tests of the rats in the experimental group were normal, and the blood and biochemical indexes were in the normal range. The hApoC Ⅲ transgenic mini-pigs were initially shown to be safe to the biological environment without adversely affecting the environment.
