目的 建立一个同时测定甘草甜素、甘草次酸血浆浓度变化监测的方法 ,进一步用于“脾主药动学”假说的验证 .方法 采用内标定量的反相高效液相色谱法 ,以甲醇 -水 -醋酸 (88∶ 11∶ 1)为流动相 ,ODS- 3柱 (15 0 mm× 4.6 mm,5μm)为固定相 ,在紫外 2 5 0 nm检测 .结果 GL,GA在此线性范围内 ,线性关系良好、精密度较高 .结论 本方法具有简便、灵敏、快速而准确的特点 ,适用于注射 GL制剂或口服中药复方前后 GL和 GA的体内、体外的药代动力学研究 ,为进一步进行中药复方的药物监测奠定了基础
Objective To establish a method for the simultaneous determination of changes in plasma concentrations of glycyrrhizin and glycyrrhetinic acid, and to further verify the hypothesis of “main pharmacokinetics of spleen.” Methods Internal standard quantification reversed-phase high performance liquid chromatography with methanol- Water-acetic acid (88:11:1) was used as the mobile phase and the ODS-3 column (15 0 mm×4.6 mm, 5 μm) was used as the stationary phase. UV detection was performed at 250 nm. The results showed that GL and GA were within this linear range. The linearity is good and the precision is high. Conclusions This method is simple, sensitive, rapid and accurate. It is suitable for in vitro and in vivo pharmacokinetic studies of GL and GA before and after injection of GL preparation or oral Chinese herbal compound for further study.
