目的:测定紫芸解毒止痛酊中的重金属和微量元素含量,并探究其镇痛作用机制。方法:利用火焰原子吸收光谱法测得紫芸解毒止痛酊重金属含量,通过小鼠致痛模型和疼痛因子的ELASA检测初步分析镇痛作用机制。结果:紫芸解毒止痛酊不含重金属镉、铅、铬、镍,含有少量的铜和锌,浓度分别为0.0185和0.56 mg/L;紫芸解毒止痛酊能缓解急性疼痛和甲醛诱发的周围炎性疼痛,且其对皮肤的刺激性小于雪上一枝蒿速效止痛搽剂;紫芸解毒止痛酊可通过抑制前列腺素E_2并上调β内啡肽含量以达到镇痛效果,但对致痛因子一氧化氮的影响较小。结论:紫芸解毒止痛酊产品重金属含量满足药物安全性要求,为其作为外用镇痛药物的使用提供理论依据。
Objective: To determine the contents of heavy metals and trace elements in Ziyunjiedu Zhitong tincture and explore the mechanism of its analgesic action. Methods: The contents of heavy metals in Ziyunjiedu Zhitong tincture were determined by flame atomic absorption spectrometry. The mechanism of analgesic action was analyzed by ELASA test in mice induced by pain model and pain factor. Results: Ziyunjiedu Zhitong tincture does not contain heavy metals such as cadmium, lead, chromium and nickel, with a small amount of copper and zinc concentrations of 0.0185 and 0.56 mg / L, respectively. Ziyunjiedu Zhitong tincture can relieve acute pain and formaldehyde-induced peripheral inflammation Pain in the skin, and its irritation to the skin is less than the quick-acting analgesic liniment on the snow; Ziyunjiedu Zhitong tincture can inhibit the prostaglandin E_2 and up-regulate the content of β-endorphin to achieve the analgesic effect, Nitrogen has less effect. Conclusion: The content of heavy metals in Ziyunjiedu Zhitong tincture can meet the requirements of drug safety and provide a theoretical basis for its use as topical analgesic drug.
