目的研究细圆藤[Pericampylus glaucus(Lam.)Merr.]的三萜类化学成分及其抗肿瘤活性。方法采用各种色谱技术分离纯化三萜类化合物,根据理化性质及波谱数据鉴定结构。采用MTT法测试三萜化合物的抗肿瘤活性。结果从细圆藤枝叶中分离鉴定了5个五环三萜类化合物,分别是hopenone-B(1)、hopenol-B(2)、22-hydroxyhopan-3-one(3)、erythrodiol3-palmitate(4)、5β,24-cyclofriedelan-3-one(5)。化合物4对K562细胞增殖有抑制作用,在100μg.mL-1质量浓度下抑制率为47%,其余化合物无明显抑制作用。结论化合物1~5为首次从细圆藤属植物中分离得到,其中4为细圆藤抗肿瘤活性成分的首例报道,其抗肿瘤活性也是首次报道。首次报道化合物2的13C-NMR谱数据以及化合物1和5的1H-NMR、13C-NMR谱数据。
Objective To study the triterpenoid chemical constituents and antitumor activity of Pericampylus glaucus (Lam.) Merr. Methods A variety of chromatographic techniques were used to separate and purify triterpenoids. Structures were identified based on physicochemical properties and spectral data. MTT assay was used to test the antitumor activity of triterpene compounds. Results Five pentacyclic triterpenoids were isolated and identified from the leaves of Cyclobalanopsis vines. They were hopenone-B (1), hadopenol-B (2), 22-hydroxyhopan-3-one (3), and erythrodiol 3-palmitate ( 4), 5β, 24-cyclofriedelan-3-one (5). Compound 4 inhibited the proliferation of K562 cells, and the inhibition rate was 47% at a concentration of 100 μg.mL-1. The other compounds had no obvious inhibitory effect. Conclusion Compounds 1~5 were isolated from the plants of the genus Cyanidia for the first time. Among them, 4 was the first report of the antitumor active components of Cynoden diphylla. Its antitumor activity was also reported for the first time. The 13C-NMR spectrum data of Compound 2 and 1H-NMR and 13C-NMR spectrum data of Compounds 1 and 5 were reported for the first time.
