本文对苯骈二氮卓类药物的热裂解机制以及其簿层展开后重氮偶合显色的色谱方法测定该类药物及其它含氮药物进行了详细研究。以氯氮 为例,利用TLC和GC-MS法,证实了其主要裂解产物之一为2-氨基-5-氯苯基苯甲酮,以及色谱显色机制为热裂解产物的重氮偶合显色。建立了用于苯骈二氮类及其它含氮药物分析的裂解薄层色谱法。展开系统S_1:氯仿-乙酸乙酯-正丁醇-浓氨水(7∶3∶0.9∶0.05)S_2:四氯化碳-乙醚-正丁醇-浓氨水(7∶3∶l.5∶0.03)。临床毒物分析实样测定结果满意。本法简便、快速、灵敏、专属,最低检测限达0.1μg,适合其它含芳伯胺类药物分析。
In this paper, the mechanism of pyrolysis of benzodiazepines and the determination of these and other nitrogen-containing drugs by diazotization and colorimetry were studied in detail. Taking chloronitrogen and nitrogen as examples, one of the major pyrolysis products of 2-amino-5-chlorophenyl benzophenone was confirmed by TLC and GC-MS, and the diazo coupling of the pyrolysis product color. A pyrolysis thin layer chromatography was developed for the analysis of benzodiazepines and other nitrogen-containing drugs. System S_1: chloroform-ethyl acetate-n-butanol-concentrated ammonia (7: 3: 0.9: 0.05) S 2: carbon tetrachloride-ether-n-butanol- .5:0.03). Clinical analysis of poison samples satisfactory results. This method is simple, rapid, sensitive and exclusive, the minimum detection limit of 0.1μg, suitable for other aromatic amine containing drugs analysis.
