为探讨淋巴细胞表面Fas及其配体(FasL)在IL-2和IL-12作用下表达的变化,以及Fas和FasL与淋巴细胞抗肿瘤活性的关系,作者用免疫荧光标记和流式细胞仪分析了IL-2和IL-12激活的杀伤细胞(IL-2AK和IL-12AK)Fas和FasL的表达;分别用乳酸脱氢酶(LDH)释放法和annexin-V-FITC标记检测IL-2AK和IL-12AK杀伤的肿瘤细胞的坏死和凋亡。结果表明:①正常人外周血淋巴细胞(PBL)(20.40±5.69)%表达Fas,但无FasL表达;3株消化道肿瘤细胞系恒定表达Fas,始终不表达FasL;②IL-2与IL-12可诱导LAK细胞表达FasL(P<0.01),并促使Fas表达显著增加(P<0.01);③LAK细胞致肿瘤细胞死亡中FasL表达与annexin-V标记阳性率和肿瘤细胞表面Fas表达正相关。由此推论,Fas-FasL途径是IL-2与IL-12诱导活化的LAK细胞杀伤肿瘤的途径之一,淋巴细胞致肿瘤凋亡分别与淋巴细胞FasL表达和肿瘤细胞Fas表达一致。
To investigate the changes of the expression of Fas and its ligand (FasL) on the surface of lymphocytes under the action of IL-2 and IL-12, and the relationship between Fas and FasL and the anti-tumor activity of lymphocytes, the authors used immunofluorescence labeling and flow cytometry The expressions of Fas and FasL in IL-2 and IL-12 activated killer cells (IL-2AK and IL-12AK) were analyzed. The expression of IL-2AK was detected by lactate dehydrogenase (LDH) release assay and annexin-V- And tumor necrosis and apoptosis of IL-12AK-killed tumor cells. The results showed that: (1) normal human peripheral blood lymphocytes (PBL) expressed Fas but no FasL expression; 3 of the gastrointestinal cancer cell lines constantly expressed Fas and never expressed FasL; ②IL-2 And IL-12 induced FasL expression in LAK cells (P <0.01), and promoted the expression of Fas significantly (P <0.01). (3) FasL expression was positively correlated with the positive rates of annexin-V in tumor cells Fas positive cell surface expression. It is concluded that Fas-FasL pathway is one of the pathways of killing tumor cells induced by IL-2 and IL-12-activated LAK cells. Lymphocyte-induced apoptosis is consistent with FasL expression of lymphocytes and Fas expression of tumor cells, respectively.
