将老年(26~27个月)及成年(3~4个月)大鼠脑切片用乙醛酸诱发单胺类荧光法,显示其黑质多巴胺(DA)。形态学观察见到,成年大鼠黑质神经元胞体充满强蓝绿色 DA 荧光,突起近端亦显荧光;而老年大鼠黑质神经元胞体蓝绿色 DA 荧光明显减弱,代之以较强的金黄色脂褐素荧光,DA 荧光细胞数量减少。显微荧光光度术对神经元 DA 荧光的定性和定量分析表明,DA 荧光发射光谱峰值为490 nm;脂褐素荧光发射光谱峰值为550 nm。成年组 DA 荧光强度((?)±S_(?))为55.813±3.032,老年组 DA 荧光强度((?)±S_(?))为28.905±2.746,P<0.001,说明老年大鼠黑质DA 浓度较成年组显著降低。本文将所得结果与已有的研究进行了对比分析,并对 DA 降低的可能影响进行了初步讨论。
The brain sections of senile (26-27 months) and adult (3-4 months) rats were induced with glyoxylate-induced monoamine fluorescence to show the substantia nigra dopamine (DA). Morphological observation showed that the substantia nigra neuronal soma of adult rats was filled with strong blue-green DA fluorescence and the proximal part of the protuberance also showed fluorescence. However, the DA blue-green fluorescence of the substantia nigra neuronal cell bodies in aged rats was significantly weakened, Golden lipofuscin fluorescence, DA fluorescent cells decreased. The qualitative and quantitative analysis of DA fluorescence of neurons by micro-fluorescence spectroscopy showed that the peak value of DA fluorescence emission was 490 nm and the peak value of lipofuscin fluorescence emission was 550 nm. The DA fluorescence intensity (± S_ (?)) In adult group was 55.813 ± 3.032, the DA fluorescence intensity (± S_ (?)) In aged group was 28.905 ± 2.746, P <0.001, indicating that the substantia nigra DA concentration was significantly lower than the adult group. In this paper, the results obtained with the existing research conducted a comparative analysis, and the possible impact of DA to reduce the initial discussion.
