目的研究胰岛β细胞损伤后胰岛再生过程中Wnt/β-catenin信号通路的生物调控。方法 STZ破坏新生大鼠胰岛β细胞,免疫组织化学观察1 d、2 d、3 d中Wnt/β-catenin通路β-catenin为核心的上、下游基因APC、c-Myc以及Nestin在胰岛再生过程中的表达。结果β-catenin在残留的胰岛细胞中表达上调,并在胰腺间质中异常表达;APC表达于胰腺导管上皮及残留的胰岛细胞;c-Myc则主要表达于胰腺腺泡上皮及胰腺间质内;Nestin表达分布于残留的胰岛细胞和胰腺导管上皮。结论 Wnt/β-catenin信号通路在胰岛再生中可能通过促进胰腺干细胞分化与增殖而调控。
Objective To study the biological regulation of Wnt / β-catenin signaling pathway during pancreatic islet cell regeneration after islet β cell injury. Methods STZ was used to injure the islet β-cells of neonatal rats. The upstream and downstream genes APC, c-Myc and Nestin were observed by immunohistochemistry at 1, 2 and 3 days after Wnt / β-catenin pathway β-catenin pathway, In the expression. Results β-catenin was up-regulated in residual islet cells and was abnormally expressed in pancreatic stroma. APC was expressed in pancreatic ductal epithelium and residual islet cells. C-Myc was mainly expressed in pancreatic acinar epithelium and pancreatic interstitium ; Nestin expression distributed in the residual islet cells and pancreatic ductal epithelium. Conclusion The Wnt / β-catenin signaling pathway may be regulated by promoting pancreatic stem cell differentiation and proliferation during pancreatic islet regeneration.
