从小球藻(Chlorella vulgaris)培养基中分离纯化到一种具有抗肿瘤活性的成分(ARS2),根据Matrix-assisted laser desorption/ionlzation(MALDI)质谱,确定ARS2为一种糖蛋白,其分子中含66.9%碳水化合物(主要是D-半乳糖)和35.2%蛋白质。采用甲基化分析和~(13)C-NMR法,进一步确定碳水化合物部分含β-1,6-D-吡喃半乳糖骨架。另外,还鉴定出蛋白质部分NH_2末端有15种氨基酸连接,并被命名为DVGEAFPTVVDAL-VA,根据肼解、过碘酸盐氧化和蛋白水解作用,确定该部分是抗肿瘤所必需的。
A component with antitumor activity (ARS2) was isolated and purified from Chlorella vulgaris medium. According to the Matrix-assisted laser desorption/ionlzation (MALDI) mass spectrometry, it was determined that ARS2 is a glycoprotein and its molecule contains 66.9% carbohydrate (mainly D-galactose) and 35.2% protein. Using methylation analysis and ~(13)C-NMR method, it was further confirmed that the carbohydrate moiety contained the [beta]-1,6-D-galactopyranoside skeleton. In addition, 15 amino acid linkages were identified at the NH 2 terminus of the protein and named DVGEAFPTVVDAL-VA, which was determined to be required for anti-tumor activity based on the hydrolysis, periodate oxidation, and proteolysis.
