目的:建立血浆中华法林对映体浓度的立体选择性高效液相色谱法。方法:血浆标本经碱洗、酸化和乙醚提后,与碳苄氧基-L-脯氨酸进行柱前衍生化为差向体。以正己烷-乙酸乙酯-乙酸-甲醇为流动相,经普通硅胶柱分离对映体和在紫外λ313nm处检测。结果:对映体标准曲线范围0~4.0mg/L,最低检出限<0.1mg/L,批内,批间平均RSD均<7%,平均回收率100.22%,临床上常用药物对本法没有干扰。结论:本法简便可靠,可用于临床华法林对映体的药动学和药效学研究。
Objective: To establish a stereoselective HPLC method for determination of warfarin enantiomers in plasma. Methods: The plasma samples were subjected to precolumn derivatization with epi-benzaldehyde, L-proline and acidified with diethyl ether, and then differentiated into epimer. The enantiomers were separated on a normal silica gel column with n-hexane-ethyl acetate-acetic acid-methanol as mobile phase and detected at λ313 nm. Results: The standard curve of enantiomers ranged from 0 to 4.0 mg / L with the minimum detectable limit of <0.1 mg / L. The mean RSD between batches and batches was <7% and the average recovery was 100.22% Medicines do not interfere with this law. Conclusion: This method is simple and reliable, and can be used to study the pharmacokinetics and pharmacodynamics of warfarin enantiomers.
