目的探讨血管内皮生长因子(VEGF)反义寡核苷酸对人类前列腺癌细胞PC-3生长及VEGF表达的影响。方法采用流式细胞术检测反义及正义寡核苷酸作用下前列腺癌细胞株PC-3 VEGF表达情况,MTT法检测VEGF反义及正义寡核苷酸对PC-3细胞生长抑制作用。结果在浓度为5~20μmol/L的反义寡核苷酸作用48 h后,肿瘤细胞VEGF表达量为0.102±0.008~0.037±0.006(P<0.05),细胞的生长抑制率为25.2%~43.9%(P<0.05),且存在浓度依赖性。结论 VEGF反义寡核苷酸可能通过抑制前列腺癌细胞PC-3 VEGF的表达而抑制肿瘤细胞的生长。
Objective To investigate the effect of vascular endothelial growth factor (VEGF) antisense oligonucleotide on the growth of human prostate cancer cell line PC-3 and the expression of VEGF. Methods Flow cytometry was used to detect the expression of PC-3 VEGF in antisense and sense oligodeoxynucleotides. The antitumor activity of VEGF antisense and sense oligodeoxynucleotides on PC-3 cells was determined by MTT assay. Results The expression of VEGF in tumor cells was 0.102 ± 0.008 ~ 0.037 ± 0.006 (P <0.05) at the concentration of 5 ~ 20μmol / L for 48 h and the growth inhibition rate was 25.2% ~ 43.9 % (P <0.05), and there is a concentration-dependent manner. Conclusion VEGF antisense oligonucleotides may inhibit the growth of tumor cells by inhibiting the expression of PC-3 VEGF in prostate cancer cells.
