为改进基于酵母朊病毒[PSI+]的抗朊病毒药物筛选模型灵敏度和促朊病毒形成条件筛选模型准确度偏低的不足,采用表型分析法研究了盐酸胍、氯化镁对4种酵母朊病毒菌株的表型影响,结果表明:ERG6Δ酵母朊病毒[PSI+]菌株对盐酸胍最为敏感,SSBΔ酵母朊病毒[psi-]菌株对氯化镁最为敏感;ERG6Δ酵母朊病毒[PSI+]菌株适合构建高灵敏度抗朊病毒药物筛选模型,SSBΔ酵母朊病毒[psi-]菌株适合构建高准确度促朊病毒形成条件筛选模型。
In order to improve the sensitivity of the prion drug screening model based on the yeast prion [PSI +] and the low accuracy of the screening model for prion production, the effects of guanidine hydrochloride and magnesium chloride on four yeast prion strains The results showed that: the strain of ERG6Δ yeast prion [PSI +] was the most sensitive to guanidine hydrochloride, and the strain of SSBΔ yeast prion [psi-] was the most sensitive to magnesium chloride; the strain of ERG6Δ yeast prion [PSI +] was suitable for construction of high sensitivity anti- Virus drug screening model, SSBΔ yeast prion [psi-] strains are suitable for constructing high-precision screening conditions for pro-prion formation conditions.
