采用分光光度法对云芝发酵浸膏中总三萜化合物含量进行测定。结果表明:5%香草醛-冰醋酸用量为0.3mL,高氯酸用量为0.8 mL,反应温度为70℃,反应时间为20 min,加冰醋酸5 mL,摇匀,在570 nm处测定其吸光值,效果最佳。标准曲线方程为Y=0.0092x-0.0447,R2=0.9991,吸光值与齐墩果酸含量在13μg~91μg范围内呈良好的线性关系;三萜类化合物的质量分数为0.564%,RSD=0.7%。齐墩果酸加样回收率98.60%~101.88%,平均加样回收率为99.93%,RSD为1.12%;在反应结束0~40 min内,吸光值相对稳定。该方法具有简便、快速、灵敏、重复性好。
The content of total triterpene in Fermented Yunzhi was determined by spectrophotometry. The results showed that the dosage of 5% vanillin-glacial acetic acid was 0.3mL, the dosage of perchloric acid was 0.8mL, the reaction temperature was 70 ℃, the reaction time was 20 min, Absorbance, the best effect. The standard curve equation is Y = 0.0092x-0.0447, R2 = 0.9991, the absorbance value and oleanolic acid content in the range of 13μg ~ 91μg showed a good linear relationship; triterpenoids mass fraction of 0.564%, RSD = 0.7% . The recovery of oleanolic acid was 98.60% ~ 101.88%, the average recovery was 99.93%, RSD was 1.12%. The absorbance was relatively stable within 0 ~ 40 min after the reaction. The method is simple, fast, sensitive and repeatable.
