目的:探讨突变的膜结合型 TNF-α基因对人胃癌细胞生长的影响.方法:用突变的 膜结合型 TNF-α基因作为目的基因,以逆转录病毒为载体将目的基因导 入人胃癌细胞 MGC803,经 G418筛选得到抗性克隆 MGCT803结果: Southern杂交证实目的基因已整合在 MGCT803细胞中.用 ELISA方法检测MGCT803细胞表面TNF- α表达量明显增 高.MGCT803细胞形态、体外增殖能力无显著改变.裸鼠体内接种表明,MGCT803细胞在裸鼠体 内的生长受到明显抑制.结论:膜结合型TNF-α基因导入人胃癌细胞后其动物体内致瘤 性明显下降.
Objective: To investigate the effect of mutant membrane-bound TNF-α gene on the growth of human gastric cancer cells. Methods: The gene was transfected into human gastric cancer cell line MGC803 by using the retroviral vector as the target gene and the resistant clone MGCT803 was screened by G418. Southern blotting confirmed that the target gene has been integrated into MGCT803 Cells. The expression of TNF-α in MGCT803 cells was significantly increased by ELISA. MGCT803 cell morphology, proliferation in vitro no significant change. In vivo vaccination of nude mice showed that the growth of MGCT803 cells in nude mice was significantly inhibited. Conclusion: The TNF-α gene of human membrane-bound TNF-α gene in human gastric cancer cells in vivo tumorigenicity decreased significantly.
