目的 研究肝细胞癌患者乙型肝炎病毒 (hepatitisBvirus,HBV)全基因组的结构及特点。方法 PCR扩增并克隆乙型肝炎病毒表面抗原 (HBsAg)阳性肝癌患者血清中HBV全基因组DNA ,测序并进行基因结构分析。结果 获得不同肝癌患者来源的 2 2株HBV全基因组DNA ,均属于C或B基因型和adr或adw2血清型。与标准株相比 ,肝癌患者来源的HBV在表面抗原、核心抗原的B/T细胞表位、X蛋白的反式激活区及增强子Ⅱ、核心启动子区发生了一些有意义的共有变异。结论HBV的基因型与基因变异可能与HBV相关性肝癌的发生发展有关。
Objective To study the structure and characteristics of whole genome of hepatitis B virus (HBV) in patients with hepatocellular carcinoma. Methods HBV whole genome DNA was amplified by PCR and cloned from the serum of hepatitis B virus surface antigen (HBsAg) -positive hepatocellular carcinoma (HCC) patients, sequenced and analyzed for gene structure. Results A total of 22 HBV genomic DNAs from different liver cancer patients were obtained, belonging to genotype C or B and adr or adw2 serotypes. Compared with the standard strains, HBV originating from HCC patients had some meaningful common mutations in surface antigen, B / T cell epitope of core antigen, transactivation region of X protein, enhancer II and core promoter region. Conclusion HBV genotypes and genetic variations may be related to the occurrence and development of HBV-related hepatocellular carcinoma.
