抑制消减杂交是最近提出的以消减杂交为基础结合抑制 PCR方法发展起来的分离克隆新基因的一种新策略。该方法具有简便易行、特异性高、背景低、重复性好、能分离出丰度较低的特异性片段等特点。本文介绍其工作原理、方法以及在肿瘤基因克隆、发育生物学、免疫调控及原核生物基因组分析等方面的应用 ,最后对方法的优缺点和应用前景作了分析
Suppression subtractive hybridization (SSH) is a new strategy recently proposed to isolate and clone new genes based on subtractive hybridization and suppression PCR. The method has the characteristics of simple and convenient, high specificity, low background, good repeatability and the ability to separate specific fragments with lower abundance. This article describes its working principle, methods and applications in tumor gene cloning, developmental biology, immune regulation and prokaryotic genome analysis, etc. Finally, the advantages and disadvantages of the method and the application prospects are analyzed
