目的:研究粘膜方是否通过上调外周B淋巴细胞Cosmc的表达,从而减少IgA肾病患者IgA1异常糖基化的量。方法:从IgA肾病患者及健康体检者采集静脉血,分离外周B淋巴细胞,采用体外细胞培养的方法,与正常人肾小球系膜细胞共培养后收集上清,与粘膜方含药血清共作用,采用实时RT-PCR和Western Blot测定Cosmc和TGF-β1mRNA和蛋白的表达水平。结果:IgA肾病患者外周血B淋巴细胞Cosmc的mRNA和蛋白表达比健康组减少,且P<0.01,差异具有统计学意义;经粘膜方含药血清共作用后,IgA肾病患者外周B淋巴细胞Cosmc的mRNA和蛋白水平表达增加,且P<0.001,差异具有统计学意义;IgA肾病患者外周血B淋巴细胞TGF-β1的mRNA和蛋白表达比健康组上升,且P<0.05,差异具有统计学意义;经粘膜方含药血清共作用后,IgA肾病患者外周B淋巴细胞TGF-β1的mRNA和蛋白表达下降,且P<0.01,差异具有统计学意义。结论:在体外人肾系膜细胞培养实验中,具有和解法立法组方的粘膜方能上调IgA肾病患者外周B淋巴细胞的Cosmc mRNA和蛋白的表达水平,并下调IgA肾病患者外周B淋巴细胞的TGF-β1和蛋白的表达水平。
OBJECTIVE: To study whether mucosal prescription reduces the IgA1 abnormal glycosylation in IgA nephropathy patients by up-regulating the expression of Cosmc in peripheral B lymphocytes. Methods: Venous blood was collected from patients with IgA nephropathy and healthy subjects. Peripheral blood B lymphocytes were isolated and cultured in vitro. After co-cultured with normal human glomerular mesangial cells, The real-time RT-PCR and Western Blot were used to detect the expression of Cosmc and TGF-β1 mRNA and protein. Results: The mRNA and protein expressions of Cosmc in B lymphocytes in patients with IgA nephropathy were significantly lower than those in healthy controls (P <0.01), and the difference was statistically significant. After co-administration of mucosal drug-containing serum, the expressions of Cosmc The mRNA and protein expression of TGF-β1 in peripheral blood B lymphocytes increased in patients with IgA nephropathy compared with healthy controls (P <0.05), and the difference was statistically significant ; After transmucosal drug-containing serum co-administration, the mRNA and protein expression of TGF-β1 in peripheral B lymphocytes decreased in patients with IgA nephropathy, with P <0.01, the difference was statistically significant. CONCLUSIONS: In vitro human mesangial cell culture experiments, the mucosa with the reconciliation protocol group can up-regulate the expression of Cosmc mRNA and protein in the peripheral B lymphocytes of patients with IgA nephropathy and down-regulate the peripheral B lymphocytes in IgA nephropathy patients TGF-β1 and protein expression levels.
