目的:探讨心肌梗死后不同时相梗死区及梗死周边区,miR-711-Sp1通路表达谱与心脏纤维化病理过程的相关性。方法:采用结扎大鼠冠状动脉左前降支方法制作大鼠心肌梗死模型,Real-time PCR法评价大鼠心脏miR-711表达水平;采用Western blot方法评价大鼠心脏Sp1蛋白表达水平;应用天狼猩红染色评价心脏纤维化病理改变。结果:大鼠心肌梗死后2d,梗死区miR-711表达无明显变化,Sp1表达下调,纤维组织减少;梗死周边区miR-711表达显著上调(P<0.01),Sp1表达无明显变化,纤维组织无明显变化。梗死后7d,梗死区miR-711表达显著下调(P<0.01),Sp1表达回升至对照水平,纤维组织开始增加;梗死周边区miR-711表达回降至对照水平,Sp1表达无明显变化,纤维组织增加不明显。梗死后14d,miR-711表达显著下调(P<0.01),Sp1表达显著上调(P<0.01),纤维组织明显增多;梗死周边区miR-711表达显著下调(P<0.05),Sp1表达显著上调(P<0.01),纤维组织沉积显著增加。结论:大鼠心肌梗死后不同区域不同时相,miR-711-Sp1及纤维化表达呈动态相关表达。
OBJECTIVE: To investigate the correlation between the miR-711-Sp1 pathway expression and the pathological process of cardiac fibrosis in infarcted and infarcted peripheral areas at different phases after myocardial infarction. Methods: The model of myocardial infarction was established by ligating the left anterior descending coronary artery in rats. Real-time PCR was used to evaluate the expression of miR-711 in rat heart. Western blot was used to evaluate the expression of Sp1 protein in rat heart. Scarlet staining for pathological changes in cardiac fibrosis. Results: The expression of miR-711 in the infarcted area was significantly increased (P <0.01), while the expression of miR-711 in the infarcted area was not significantly changed at the 2nd day after myocardial infarction No significant changes. At 7 days after infarction, the expression of miR-711 in infarcted area was significantly down-regulated (P <0.01), and the expression of Sp1 was up to the level of control. The expression of miR-711 in peripheral area of infarcted area was decreased to the level of control. Tissue increase is not obvious. On the 14th day after infarction, the expression of miR-711 was significantly downregulated (P <0.01), the expression of Sp1 was significantly up-regulated (P <0.01), and the expression of miR-711 was significantly decreased (P <0.05) (P <0.01), fibrous tissue deposition increased significantly. CONCLUSION: The expression of miR-711-Sp1 and fibrosis in different regions of myocardium at different time points after myocardial infarction were dynamically correlated.
