目的:研究云南松松塔提取物含药血清对乙醇损伤大鼠肝细胞的抗氧化作用进行研究。方法:取SD大鼠10只,随机分为正常组和云南松松塔提取物组(1 g·kg-1),每组5只,每天ig 2次,连续3 d,于末次ig 8 h后制备含药血清。取新生大鼠处死,无菌条件下,取肝脏分离正常肝细胞进行培养,将预培养的肝细胞分为正常组、模型组、空白组、3个不同质量分数云南松松塔提取物含药血清组(25%,50%,100%),用不同质量分数的云南松松塔提取物含药血清对体外培养的大鼠正常肝细胞进行预处理,1 h后,除正常组,空白组外,其余组加入100 mmol·L-1乙醇诱导损伤,继续培养8 h后,MTT法检测大鼠肝细胞存活率,酶标法检测培养液中丙氨酸氨基转移酶(ALT),超氧化物歧化酶(SOD)活性和丙二醛(MDA),还原型谷胱甘肽(GSH)含量。结果:与正常组比较,受损大鼠肝细胞经5%和10%含药血清处理后,存活率明显增大,细胞培养液中ALT活性和MDA含量明显降低,SOD活性和GSH含量明显升高(P<0.05,P<0.01)。结论:云南松松塔提取物含药血清对乙醇损伤大鼠肝细胞有一定的抗氧化作用。
OBJECTIVE: To study the antioxidative effect of drug-containing serum from pine cone tower on the hepatocytes in ethanol-injured rats. Methods: Ten Sprague-Dawley rats were randomly divided into normal group and Pinus yunnanensis extract group (1 g · kg-1), 5 rats in each group, ig twice a day for 3 consecutive days. After preparation of drug-containing serum. Neonatal rats were sacrificed, and under normal conditions, normal liver cells were isolated from the liver and cultured. The pre-cultured hepatocytes were divided into normal group, model group and blank group, and three different concentrations of Pinus yunnanensis extract Serum group (25%, 50%, 100%), with different mass fraction of Yunnan pine pineapple extract containing serum on rat normal liver cells were pretreated 1 h later, except the normal group, the blank group , The other groups were treated with 100 mmol·L -1 ethanol to induce injury. After cultured for 8 h, the survival rate of rat hepatocytes was detected by MTT assay. The levels of alanine aminotransferase (ALT), superoxide (SOD) activity, malondialdehyde (MDA) and reduced glutathione (GSH) contents were determined. Results: Compared with the normal group, the survival rate of injured rat hepatocytes was significantly increased after treatment with 5% and 10% serum, the ALT activity and MDA content in the cell culture fluid decreased significantly, and the SOD activity and GSH content increased significantly High (P <0.05, P <0.01). CONCLUSION: The drug-containing serum of Pinus yunnanensis L. extract has certain antioxidative effect on hepatocytes of ethanol-injured rats.
