[目的]探讨hOGG1Ser326Cys多态与食管癌易感性、临床病理特性之间的关系。[方法]选取河南省新乡市188例食管癌患者和203例正常对照作为研究对象,采用病例-对照研究方法,应用多聚酶链反应-单链构象多态(polymerase chain reaction-single strand conformation polymorphism,PCR-SSCP)技术,分析两组人群hOGG1基因第326位点Ser/Ser、Ser/Cys和Cys/Cys基因型的分布,以及各基因型在不同临床病理特性中的分布差异。[结果]hOGG1三种基因型(Ser/Ser、Ser/Cys和Cys/Cys)在两组人群中的分布无统计学差异(χ2=2.13,P=0.344);在食管鳞状细胞癌组织学分型中,Ⅲ级与Ⅰ级相比,hOGG1Cys/Cys基因型在分化程度差的食管癌中的分布明显增加(OR=3.69,95%CI:1.16 ̄11.76)。[结论]hOGG1Cys/Cys基因型与食管鳞状细胞癌组织低分化相关,提示hOGG1活性降低可能预示食管癌预后不良。
[Objective] To investigate the relationship between hOGG1Ser326Cys polymorphism and the susceptibility and clinicopathological features of esophageal cancer. [Method] A total of 188 esophageal cancer patients and 203 normal controls in Xinxiang City of Henan Province were selected as the research objects. By case-control study, polymerase chain reaction-single strand conformation polymorphism -SSCP) technique was used to analyze the distribution of Ser / Ser, Ser / Cys and Cys / Cys at the 326th locus of hOGG1 gene and the distribution of different genotypes in different clinicopathological features. [Results] There was no significant difference in the distribution of the three genotypes of hOGG1 (Ser / Ser, Ser / Cys and Cys / Cys) between the two groups (χ2 = 2.13, P = 0.344) In type Ⅲ, the distribution of hOGG1Cys / Cys genotypes was significantly higher in poorly differentiated esophageal cancer than in grade Ⅲ (OR = 3.69, 95% CI: 1.16-11.76). [Conclusion] The hOGG1Cys / Cys genotype is associated with the poorly differentiated esophageal squamous cell carcinoma, suggesting that the decrease of hOGG1 activity may predict the poor prognosis of esophageal cancer.
