利用葡萄糖和果糖在碱性条件下烯醇化反应速度不同,提出了速差动力学光度法同时测定葡萄糖和果糖的新方法。通过实验条件的选择,确定了葡萄糖和果糖与斐林试剂反应,与磷钼酸显色的动力学反应级数为零级;且反应表观速率常数分别为:0.108mg/L·min,0.427mg/L·min;测量浓度的线性范围均为:2~8mg/L;最低检测限为1.0×10-9g/L;对人工合成样品测定结果的相对标准偏差为1.20%~341%;对实际样品测定结果的标准回收率为95%~102%。
Based on the different enolase reaction rates of glucose and fructose under alkaline conditions, a new method of simultaneous determination of glucose and fructose by differential kinetic spectrophotometry was proposed. Through the selection of experimental conditions, the kinetic reaction order of glucose and fructose reacted with Fehling reagent was determined to be zero order. The apparent rate constants of reaction were 0.108mg / L · min, 0.427mg / L · min. The linear range of the measured concentration was 2 ~ 8mg / L, the minimum detection limit was 1.0 × 10-9g / L. The relative standard deviation of the results of synthetic samples was 1.20 % ~ 341%; The standard recovery of the actual sample was 95% ~ 102%.
