目的:检测全反式维甲酸(ATRA)诱导后HL-60细胞核干细胞因子(NS)mRNA表达的变化,探讨NS mRNA表达与细胞分化之间的关系。方法:从NS基因的3个变异体中筛选出共同序列设计特异性引物,用终浓度为0.1μmol/L、1μmol/L、10μmol/L的ATRA诱导分化HL-60白血病细胞48h,以不加ATRA的HL-60细胞作为对照,应用逆转录-聚合酶链式反应(RT-PCR)检测HL-60细胞NS mRNA表达水平的变化。结果:ATRA诱导HL-60细胞分化后NS mRNA表达均有所降低,0.1μmol/L、1μmol/L、10μmol/L浓度下NS mRNA表达量分别下降3.72%±0.84%、58.70%±4.98%和61.60%±2.26%。其中1μmol/L和10μmol/L组与对照组和0.1μmol/L组比较,差异均有统计学意义(P均<0.05)。结论:NS可能与白血病细胞分化有关,参与调控细胞分化过程。
Objective: To detect the change of nuclear stem cell factor (NS) mRNA expression in HL-60 cells induced by all-trans retinoic acid (ATRA) and to explore the relationship between NS mRNA expression and cell differentiation. METHODS: Specific primers designed by common sequence were screened from three variants of NS gene. HL-60 leukemia cells were induced to differentiate with ATRA of 0.1μmol / L, 1μmol / L, 10μmol / L for 48h, ATRA HL-60 cells as a control, reverse transcription-polymerase chain reaction (RT-PCR) detection HL-60 cells NS mRNA expression changes. Results: The mRNA expression of NS in HL-60 cells was decreased after ATRA treatment. The expression of NS mRNA decreased by 3.72% ± 0.84% and 58.70% ± 4.98% at 0.1μmol / L, 1μmol / L and 10μmol / L, 61.60% ± 2.26%. There was significant difference between 1μmol / L and 10μmol / L groups and control group and 0.1μmol / L group (all P <0.05). Conclusion: NS may be related to the differentiation of leukemia cells and participate in the regulation of cell differentiation.
