目的研究肝细胞癌组织及其癌旁非癌肝组织中Fas/FasL(Fas ligand)的表达情况及其与临床病理的关系。方法应用免疫组化Envision和RT-PCR双重方法检测30例原发性肝细胞癌(hepatocellular carcinoma,HCC)患者的癌组织及其癌旁非癌肝组织的Fas蛋白和FasL的mRNA及其蛋白的表达情况。结果 12例(12/30,40.0%)HCC表达Fas蛋白,明显低于癌旁组织(21/30,70.0%,P<0.05).21例(21/30,70.0%)HCC的FasL蛋白为阳性,略高于癌旁组织(17/30,56.7%),两者无显著性差别,但HCC的FasL mRNA水平较癌旁组织为高(70.1/32.2,P<0.05),且同一癌组织多呈Fas FasL’表型(16/30,53.3%),癌组织Fas与FasL表达显著相关。HCC中Fas蛋白的表达与其组织学分级、癌周侵袭程度及肿瘤包膜有关(1/3/7/1,4/12,1/4/7,P<0.05).而FasL蛋白的表达与其临床病理指标无明显关系(0/2/16/3,7/21,5/13/3,P>0.05).结论肝癌可能通过下调Fas、增强FasL的表达实现免疫逃避;而Fas的表达对辅助判断肝癌的侵袭转移乃至预后可能有重要意义。
Objective To investigate the expression of Fas ligand (Fas ligand) in hepatocellular carcinoma and its adjacent non-cancerous liver tissues and its relationship with clinicopathological features. Methods The expressions of Fas and FasL mRNA and protein in 30 cases of primary hepatocellular carcinoma (HCC) and their adjacent non-cancerous liver tissues were detected by immunohistochemical Envision and RT-PCR methods. Express the situation. Results 12 cases (12/30, 40.0%) HCC expressed Fas protein, which was significantly lower than that in paracancerous tissues (21/30, 70.0%, P <0.05). FasL protein in 21 cases (17/30, 56.7%). There was no significant difference between the two groups (P> 0.05). However, the level of FasL mRNA in HCC was higher than that in paracancer tissues (70.1 / 32.2, P <0.05) Mostly showed Fas FasL phenotype (16/30, 53.3%). The expression of Fas and FasL in cancerous tissues was significantly correlated. The expression of Fas protein in HCC was correlated with histological grade, degree of invasion of cancer and tumor capsule (1/3/7 / 1,4 / 12,1 / 4/7, P <0.05) (P <0.05) .ConclusionHepatocarcinoma may achieve immune evasion by down-regulating Fas and enhancing the expression of FasL, while the expression of Fas Assisted determination of invasion and metastasis of liver cancer and prognosis may be of great significance.
