目的:采用高效液相色谱(HPLC)法建立一种同时测定金英黄归汤中芦丁、槲皮素和木犀草素的含量。方法:采用Hypersil ODS2 C18色谱柱(150 mm×4.6 mm,5μm);以甲醇-0.4%磷酸(40∶60)为流动相;检测波长为360 nm;柱温为40℃和;流速为0.8 m L·min-1。结果:芦丁、槲皮素和木犀草素分别在350.00μg·m L-1~21.88μg·m L-1(R2=0.999 8)、245.00μg·m L-1~15.31μg·m L-1(R2=0.999 9)和240.00μg·m L-1~15.00μg·m L-1(R2=0.999 9)范围内线性关系良好。中间精密度试验RSD分别为1.05%、1.16%和0.50%;稳定性试验RSD分别为2.00、%1.79%和1.60%;重复性试验RSD分别为1.78%、1.23%和1.84%;平均回收率分别为芦丁99.08%(RSD=0.81%),槲皮素100.5%(RSD=1.82%),木犀草素99.92(RSD=1.19%)。结论:建立的该测定方法简便快捷,准确可靠,重复性好,可用于同时测定金英黄归汤中芦丁、槲皮素和木犀草素的含量测定。
Objective: To establish a simultaneous determination of rutin, quercetin and luteolin in Jinying Huanggui Decoction by high performance liquid chromatography (HPLC). METHODS: Hypersil ODS2 C18 column (150 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of methanol-0.4% phosphoric acid (40:60), the detection wavelength was 360 nm, the column temperature was 40 ℃ and the flow rate was 0.8 m L · min-1. Results: The results showed that the concentrations of rutin, quercetin and luteolin were in the ranges of 350.00μg · m L-1 ~ 21.88μg · m L-1 (R2 = 0.999 8), 245.00μg · m L-1 ~ 15.31μg · m L- 1 (R2 = 0.999 9) and 240.00μg · m L-1 ~ 15.00μg · m L-1 (R2 = 0.999 9). RSD of the intermediate precision test were 1.05%, 1.16% and 0.50% respectively; RSD of stability test was 2.00%, 1.79% and 1.60% respectively; RSD of repeatability test were 1.78%, 1.23% and 1.84% 99.08% (RSD = 0.81%) of rutin, 100.5% of quercetin (RSD = 1.82%) and 99.92% of luteolin (RSD = 1.19%). Conclusion: The established method is simple, rapid, accurate, reliable and reproducible. It can be used for the simultaneous determination of rutin, quercetin and luteolin in Jinying Huanggui Decoction.
