目的 探讨支原体感染与胃癌的相关性以及胃癌细胞感染支原体后基因表达的变化。方法 建立组织芯片制备体系 ,制备包含 10 5例胃癌组织、10 1例胃癌手术切缘正常组织和 6 2例胃良性疾患的组织芯片。以猪鼻支原体特异抗体PD4为一抗 ,利用免疫组织化学的方法对组织芯片进行染色 ,判断猪鼻支原体感染的分布情况。借助基因芯片分析人胃癌细胞系MGC80 3细胞被支原体感染后基因表达水平的变化。结果 被测标本中 ,5 4 1% (5 3 98)的胃癌组织存在猪鼻支原体感染。胃癌标本正常切缘组织感染率 5 1 7(45 87)。而胃良性疾患组织感染率仅为 15 8% (9 5 7) ,与胃癌组织相比差异有显著意义 (P =0 0 0 1)。感染率与肿瘤细胞分化程度成正比。人胃癌细胞MGC80 3细胞在感染支原体后 ,在被检测的 4 80 0 0个基因位点中共有 4 0 9个基因的表达水平发生明显改变。部分凋亡、细胞粘附相关基因的表达受到抑制。结论 胃癌组织中猪鼻支原体的感染率明显高于胃良性疾患组织 ,提示猪鼻支原体感染与胃癌的发生发展可能存在一定的相关性。猪鼻支原体的感染明显影响胃癌细胞的基因表达 ,提示猪鼻支原体的感染可能会影响胃癌细胞的生物学行为。
Objective To investigate the correlation between mycoplasma infection and gastric cancer and the changes of gene expression in gastric cancer cells infected with mycoplasma. Methods Tissue microarray system was established to prepare tissue microarray including 105 cases of gastric cancer, 101 cases of gastric cancer and 4 cases of stomach benign disease. Mycoplasma hyorhinis specific antibody PD4 as the primary antibody, the use of immunohistochemical staining of tissue chips to determine the distribution of Mycoplasma hyorhinis infection. Using gene chip analysis of human gastric cancer cell line MGC80 3 cells were infected with mycoplasma gene expression levels. Results In the specimens tested, 541% (5398) of gastric cancer tissues were infected with M. hyopneumoniae. The infection rate of normal marginal tissue of gastric cancer was 5 1 7 (45 87). The infection rate of stomach benign disease was only 15 8% (957), which was significantly different from that of gastric cancer (P 0 01). Infection rate is proportional to the degree of tumor cell differentiation. After infected with mycoplasma, the expression level of 4 0 9 genes in 4 80 0 0 loci of human gastric cancer cell MGC80 3 cells changed significantly. Part of apoptosis, cell adhesion-related gene expression was inhibited. Conclusion The infection rate of Mycoplasma hyopneumoniae in gastric cancer tissues is significantly higher than that in gastric benign disease tissues, suggesting that Mycoplasma hyorhinis infection may have a certain correlation with the occurrence and development of gastric cancer. Mycoplasma hyorhinis infection significantly affect the gastric cancer cell gene expression, suggesting that infection with Mycoplasma hyorhinis may affect the biological behavior of gastric cancer cells.
