目的 :探讨衰老对大鼠脂肪组织FABP4表达改变的影响及可能机制。方法 :10只SD成年大鼠(4个月)和10只SD老年大鼠(22个月)均给予普通饮食饲养。取大鼠脂肪组织,采用q RT-PCR检测FABP4和DNMT1 mRNA表达水平,Western blot检测FABP4和DNMT1蛋白表达水平,采用巢式降落式甲基化特异性PCR(n MS-PCR)检测FABP4 DNA甲基化水平的改变。结果:与成年大鼠比较,老年大鼠脂肪组织FABP4 mRNA和蛋白表达水平分别下降了70%和68%(P<0.01),而老年大鼠脂肪组织中FABP4 DNA甲基化水平升高了0.2倍(P<0.05),DNMT1 mRNA和蛋白表达水平分别升高了1.73、1.55倍(P<0.05)。结论:衰老可引起大鼠脂肪组织FABP4表达降低,且FABP4 DNA甲基化程度升高,DNMT1表达升高可能参与其甲基化改变的调控过程。
Objective: To investigate the effect of aging on the expression of FABP4 in adipose tissue of rats and its possible mechanism. Methods: Ten SD adult rats (4 months) and 10 SD rats (22 months) were given normal diet. FABP4 and DNMT1 mRNA expression levels were detected by q RT-PCR, FABP4 and DNMT1 protein levels were detected by Western blot, and FABP4 DNA methylation level was detected by nested-type-specific methylation-specific PCR (n MS-PCR) Changes in the level of the Results: Compared with adult rats, the expression of FABP4 mRNA and protein in adipose tissue of adult rats decreased by 70% and 68% (P <0.01), while the levels of FABP4 DNA methylation in adipose tissue of elderly rats increased by 0.2 (P <0.05). The mRNA and protein expressions of DNMT1 increased by 1.73 and 1.55 folds (P <0.05). CONCLUSION: Aging can cause the decrease of FABP4 expression in adipose tissue and the increase of DNA methylation in FABP4. The increased expression of DNMT1 may be involved in the regulation of methylation changes.
