目的研究十全大补汤含药血清对人肺腺癌顺铂(DDP)敏感细胞株A549和耐药细胞株A549/DDP耐药性及凋亡的影响。方法将24只SD大鼠随机分为十全大补汤含药血清低剂量组(1.625 g/ml)、中剂量组(3.25 g/ml)、高剂量组(6.5 g/ml)、空白血清组(等体积生理盐水),每天灌胃给药1次,连续7 d后提取血清;MTT法检测各组细胞对DDP的半数抑制浓度(IC50)值,并计算耐药指数;AnnexinⅤ/PI法检测各组细胞的早期凋亡率。结果 A549细胞中,中、高剂量组IC50值均较空白血清组下降(均P<0.01),而中剂量组的IC50值与高剂量组无差别,但较低剂量组显著降低(P<0.05);A549/DDP细胞中,低、中、高剂量组的IC50值均较空白血清组下降(均P<0.01),而中剂量组IC50值与高剂量组无差别,但较低剂量组显著降低(P<0.01);DDP处理A549细胞后,中、高剂量组分别增加细胞的早期凋亡率(P<0.05,P<0.01),中、高剂量组之间无差别(P>0.05),但中剂量组较低剂量组增加细胞的早期凋亡率(P<0.05);DDP处理A549/DDP细胞后,低、中、高剂量组均增加细胞的早期凋亡率(P<0.05,P<0.01),但中、高剂量组之间无差别,而中剂量组较低剂量组增加细胞的早期凋亡率(P<0.05)。结论十全大补汤含药血清对A549及A549/DDP细胞均有降低DDP耐药性的协同增效作用,可能与其促进肿瘤细胞凋亡有关;在A549/DDP细胞中十全大补汤含药血清的协同增效作用更强,且中剂量含药血清效率最高、效果最佳。
Objective To study the effect of Shiquan Dabu Tang containing serum on drug resistance and apoptosis of human lung adenocarcinoma cell line DDP-sensitive A549 and drug-resistant cell line A549 / DDP. Methods Twenty-four Sprague-Dawley rats were randomly divided into three groups: low dose group (1.625 g / ml), medium dose group (3.25 g / ml), high dose group (Equal volume of normal saline) was given intragastrically once a day for 7 days and the serum was extracted. The half inhibitory concentration (IC50) value of DDP in each group was determined by MTT method and the drug resistance index was calculated. The AnnexinⅤ / PI method The early apoptotic rate of each group was detected. Results The IC50 values of A549 cells were lower than those of blank serum group (all P <0.01), while the IC50 values of middle dose group were not significantly different from those of high dose group (P <0.05) ). The IC50 values of A549 / DDP cells in low, medium and high dose groups were lower than those in blank serum group (all P <0.01), while the IC50 values in middle dose group were not different from those in high dose group (P <0.01). After treated with DDP, the cell apoptosis rates of A549 cells were increased (P <0.05, P <0.01), but no difference was found between the medium and high dose groups (P> 0.05) (P <0.05). However, the cell apoptosis rates of A549 / DDP treated with DDP were significantly increased (P <0.05, P <0.05) P <0.01). However, there was no difference between the medium and high dose groups, while the middle dose group and the low dose group increased the early apoptosis rate (P <0.05). Conclusions The serum containing Shiquan Dabu Tang can reduce the synergistic effect of DDP on A549 and A549 / DDP cells, which may be related to the promotion of tumor cell apoptosis. In Shijiazhuang Dabu Tang containing A549 / DDP cells Synergistic effect of drug serum stronger, and the highest dose of serum containing the highest efficiency, the best.
