目的探讨补阳还五汤联合骨髓间充质干细胞(MSCs)移植对脑缺血损伤的神经保护机制。方法将48只SD大鼠随机分为假手术组、模型组、MSCs移植组(MSCs组)、补阳还五汤联合MSCs移植组(联合组),每组12只。建立局灶性脑缺血再灌注模型,MSCs组、联合组从尾静脉注入MSCs,联合组并给予补阳还五汤,给药10天后检测碱性成纤维细胞生长因子(bFGF)基因表达情况。结果 MSCs组与联合组均能上调大鼠脑组织bFGFmRNA表达水平,与模型组和假手术组相比,差异均有统计学意义(P<0.05),联合组表达明显高于MSCs组(P<0.05)。结论补阳还五汤联合MSCs移植能通过上调脑组织bFGFmRNA表达水平,促进内源性神经营养因子bFGF基因表达而起对脑缺血再灌注损伤起保护作用。
Objective To investigate the neuroprotective mechanism of Buyang Huanwu Decoction combined with bone marrow mesenchymal stem cells (MSCs) transplantation on cerebral ischemia injury. Methods 48 SD rats were randomly divided into sham operation group, model group, MSCs transplantation group (MSCs group), Buyang Huanwu Decoction combined with MSCs transplantation group (combined group), 12 rats in each group. The model of focal cerebral ischemia-reperfusion was established. MSCs group and combination group were injected with MSCs from caudal vein, and the combination group was given BYHWD for 10 days. The expression of basic fibroblast growth factor (bFGF) . Results Both MSCs group and combined group could up-regulate the expression of bFGFmRNA in rat brain tissue. Compared with model group and sham operation group, the difference was statistically significant (P <0.05), and the expression of bFGF in combination group was significantly higher than that in MSCs group (P < 0.05). Conclusion Buyang Huanwu Decoction combined with MSCs transplantation can protect cerebral ischemia-reperfusion injury by up-regulating the expression of bFGFmRNA in brain tissue and promoting the expression of bFGF gene of endogenous neurotrophic factor.
