目的:探讨桑菊清解汤对急性吸烟大鼠氧化与抗氧化系统的影响。方法:将50只雄性大鼠,按体重随机分为正常对照组、模型组、NAC(乙酰半胱氨酸颗粒)组及高、低剂量桑菊清解汤组,每组10只。正常对照组、模型组灌胃蒸馏水;高、低剂量组桑菊清解汤灌胃相应剂量桑菊清解汤,均按1mL/100g体重给药;NAC组将NAC以蒸馏水溶解,配制成浓度为0.2g/mL的溶液,按200mg/kg体重灌胃给药。除正常对照组外,其他各组每天空腹灌胃1次后进行被动吸烟造模,连续7天。第7天后取血浆及肺泡灌洗液(BALF),检测还原型谷胱甘肽(GSH)含量及测定血清超氧化物歧化酶(SOD)活性。结果:与正常对照组比较,模型组BALF、血浆中GSH含量及BALF和血清中SOD活性均低于正常对照组(P<0.01);与模型组比较,低剂量桑菊清解汤组BALF中GSH含量及BALF和血清中SOD活性升高(P<0.05),而高剂量桑菊清解汤组和NAC组BALF、血浆中GSH含量及BALF和血清中SOD活性均升高(P<0.01)。高、低剂量桑菊清解汤对急性被动吸烟损伤动物模型气道有一定保护作用且其作用呈一定的剂量依赖关系。结论:桑菊清解汤对被动吸烟所致大鼠急性肺损伤有一定的保护作用,且其保护作用可能与抑制自由基的产生、减少脂质过氧化有关。
Objective: To investigate the effect of Sang Ju Qing Jie Tang on the oxidation and anti-oxidation system in acute cigarette smoking rats. Methods: Fifty male rats were randomly divided into normal control group, model group, NAC (acetylcysteine granules) group and high and low dose Sheng Ju Qing Jie Tang group, with 10 rats in each group. The normal control group and model group were fed with distilled water. The high and low dose groups were administrated with the corresponding dose of Sangjuqing Decoction by Sangjuqing Decoction, and the dosage was 1mL / 100g body weight. The NAC group was dissolved in distilled water to make the concentration 0.2g / mL solution, administered by 200mg / kg body weight gavage. In addition to the normal control group, the other groups were fed with fasting gas by fasting once a day for 7 consecutive days. After 7 days, plasma and alveolar lavage fluid (BALF) were taken, the content of reduced glutathione (GSH) and the activity of serum superoxide dismutase (SOD) were measured. Results: Compared with the normal control group, the content of GSH in BALF and plasma and the activity of SOD in serum in BALF and serum in model group were lower than those in normal control group (P <0.01). Compared with model group, (P <0.05). The levels of GSH in BALF, plasma and SOD activity in BALF and serum of high-dose Sangjuqingjie decoction group and NAC group were significantly increased (P <0.01) . High and low dose of Sangjuqingjie decoction has certain protective effect on the airway of acute passive smoking-induced injury in animal model and its effect has certain dose-dependent relationship. Conclusion: Sangjuqingjie decoction can protect rat acute lung injury induced by passive smoking, and its protective effect may be related to inhibiting the production of free radicals and reducing lipid peroxidation.
